透射电镜在本科实验教学中的应用

摘要 透射电镜是一种电子光学仪器,用来观察生物组织、细胞以及细胞器的超微结构,具有高分辨率、高放大倍数和直观性等特点,适用于生物、化学、材料等很多领域。以玉米“登海605”为实验材料,在本科生实验教学中利用透射电镜观察玉米叶片细胞中叶绿体的超微结构,要求学生掌握透射电镜的原理、样品制备及使用方法,更好地了解透射电镜在生物、材料等学科中的应用,培养学生理论知识与实际应用相结合的能力。 Transmission electron microscope(TEM)is an electronic optical instrument known for its high resolution,magnification and intuitiveness.It has found wide application in diverse fields such as biology,materials science among others.In this article,Maize"Denghai605"was used as the experimental material to enable undergraduate students to use TEM to observe the ultrastructure of organelles such as chloroplasts in maize leaf cells.The aim of the experiment was to enable the students to master the principles and methods of sample preparation,as well as the utilization of a TEM.This was intended to cultivate the students'ability to apply theoretical knowledge to practical situations,while at the same time enabling them to understand the application of TEM in various fields such as biology and materials science.
