剪法及温湿度对钙果4号试管苗增殖效应的影响

摘要 钙果4号试管苗在同一配方培养基继代多次后容易出现种苗老化现象。利用钙果4号试管苗茎切段腋芽萌发成梢的特性,剪掉苗木中上部茎段,对剩留的剪切苗进行保护处理,剪切时注意从基部往上最多预留2个腋芽,然后放置于湿度为65%~73%、温度为29±1℃的环境下继代培养。结果显示,腋芽可萌发出2.8个嫩梢,嫩梢长度约为4.69 cm,嫩梢上着生叶片的叶面积、叶绿素含量、比叶重等指标良好,无黄化、老化现象。结果表明,通过调整剪切方式、温湿度等因素,可减缓种苗老化,为下次继代培养提供合格材料。 Cerasus humilis cv.Four plantlets will occur seedling aging phenomenon constantly when subcultured on a same formula medium many times,fully utilize the specificity of germinated shoots from axillary buds which Cerasus humilis cv.Four stem segment plantlets subcultured.Share off the upper part stem segment,then take protective measure on the remaining cutting seedlings,including reserve 2 axillary buds from downslope on upslope when inoculated and sheared,and then subcultured on the culture shelf which set the humidity and temperature are 65~73%and 29±1℃respectively.The axillary buds can germinate bright and shaped shoots,the number of shoots is above 2.8,the length of shoots is above 4.69 cm,other measurement indicators such as the leaf area,the content of chlorophyll and the leaf specific weight represent excellent performance.Furthermore,there is no apparent seedling aging and etiolation.Through adjusting factors of shearing mode and humiture,mitigate the trend of seedling aging,and supply eligible inoculated plantlets for next subculture.
