摘要
铁死亡(Ferroptosis),一种铁依赖的,区别于凋亡、坏死、自噬的新型细胞程序性死亡。铁死亡在器官损伤和组织退行性变,以及抑制肿瘤细胞增殖中发挥重要作用。目前,利用铁死亡诱导杀灭癌细胞已成为一种新的癌症治疗方法。通过药理调控诱导和抑制铁死亡,在治疗耐药肿瘤、缺血性器官损伤和其他与广泛脂质过氧化相关的退行性疾病方面有着巨大潜能。铁死亡起效迅速,不受细胞膜上特定蛋白介导的多耐药性影响,无细胞凋亡抑制,因此可用于研发肿瘤治疗制剂。纳米材料诱导铁死亡被认为是一种基于纳米材料的工程化治疗剂新策略,以实现对各种癌症高效治疗。本次综述主要介绍癌细胞铁死亡的意义,几种癌细胞铁死亡诱导型纳米材料,以及铁死亡诱导型纳米材料在治疗癌症中的应用和纳米材料诱导癌细胞铁死亡的优势和展望。
Ferroptosis,an iron-dependent novel form of programmed cell death distinct from apoptosis,necrosis,and autophagy,plays a crucial role in organ damage,tissue degeneration,and inhibiting the proliferation of tumor cells.Currently,inducing cell death through ferroptosis has emerged as a new approach in cancer treatment.Pharmacologically regulating ferroptosis,both inducing and inhibiting,holds immense potential in treating drug-resistant tumors,ischemic organ damage,and other degenerative diseases associated with widespread lipid peroxidation.Ferroptosis acts rapidly,is not affected by multidrug resistance mediated by specific proteins on the cell membrane,and is free from apoptosis inhibition,making it suitable for the development of anticancer formulations.Nanomaterial-induced ferroptosis is considered a novel strategy in engineered therapeutic agents based on nanomaterials,aiming to achieve efficient treatment for various cancers.This review primarily discusses the significance of ferroptosis in cancer cells,various nanomaterials inducing ferroptosis,and the application of ferroptosis-inducing nanomaterials in cancer treatment.Additionally,it explores the advantages and prospects of nanomaterial-induced ferroptosis in cancer cells.
