光调控红色红曲菌M7及MrVeA基因相关突变株的功能分析

摘要 为了探讨光调控对红色红曲菌(Monascus ruber)M7、MrVeA基因相关突变株ΔMrVeA、OEMrVeA功能的影响,采用MrVeA基因敲除、过表达技术分别构建基因缺失菌株(ΔMrVeA)和过表达菌株(OEMrVeA)。以避光(黑暗)为对照,分析红光和白光对红色红曲菌M7、MrVeA基因突变株ΔMrVeA、OEMrVeA生长发育、红曲色素(Mps)和桔霉素(CIT)产生的影响。结果表明,黑暗条件下,突变株OEMrVeA闭囊壳和分生孢子的产量增加,菌丝发育、菌落颜色、生物量及色素含量与红色红曲菌(M.ruber)M7相比均无显著性差异,但产桔霉素能力比菌株M7降低7.03%,说明MrVeA基因的过表达会促进菌株M7的有性/无性繁殖,抑制桔霉素的生成。红光有利于突变株OEMrVeA的生长发育、有性/无性繁殖、红曲色素的生成,不利于突变株OEMrVeA生成桔霉素;白光不利于突变株OEMrVeA的生长发育、有性/无性繁殖、红曲色素的生成。研究结果可为低产甚至不产桔霉素的红曲菌工业化应用提供参考。 In order to investigate the effect of light control on the function of Monascus ruber M7 and MrVeA gene related mutantΔMrVeA,OEMrVeA,the gene deletion strain(ΔMrVeA)and overexpression strain(OEMrVeA)were constructed by MrVeA gene knockout and overexpression techniques,respectively.Using lucifuge(dark)as control,the effects of red light and white light on the growth and development,and the production of Monascus pigment(Mps)and citrinin(CIT)of M.ruber M7,MrVeA gene mutant strainsΔMrVeA and OEMrVeA were analyzed.The results showed that the production of cleistothecium and conidium of mutant OEMrVeA increased under dark conditions,and there was no significant difference in mycelium development,colony color,biomass and pigment content compared with M.ruber M7,but the citrinin production capacity decreased by 7.03%compared with M.ruber M7,indicating that the overexpression of MrVeA gene could promote the sexual/asexual reproduction of strain M.ruber M7 and inhibit the citrinin formation.Red light was conducive to the growth and development of the mutant OEMrVeA,sexual/asexual reproduction and Mps generation,but was not conducive to citrinin generation of mutant strain OEMrVeA.White light was not conducive to the growth and development of mutant OEMrVeA,sexual/asexual reproduction,and Mps generation.The results could provide a reference for the industrial application of Monascus with low or even no citrinin production.
