摘要
为验证NtJAZ15对烟草腺毛发生的影响,探究NtJAZ15调控腺毛发生的分子机制,本研究从K326中克隆了NtJAZ15基因,并对其进行了生物信息学和组织表达模式分析,通过创制NtJAZ15敲除和过表达突变体,分析其对腺毛发生的调控作用。结果表明,NtJAZ15-1和NtJAZ15-2在不同烟草品种中高度保守,具有典型的TIFY和Jas结构域,属于JAZ家族。NtJAZ15在各组织中均表达,且各组织中NtJAZ15-2表达量均高于NtJAZ15-1。NtJAZ15基因敲除突变体烟叶长柄和短柄腺毛密度增加,过表达突变体则相反。qRT-PCR结果表明NtJAZ15可显著影响NtWo、NtCycB2、NtHD9、NtHD12的表达,但对NtMIXTA的表达影响不显著。因此,NtJAZ15负调控烟草长柄和短柄腺毛发生,NtWo、NtCycB2、NtHD9、NtHD12可能参与NtJAZ15调控烟草腺毛发生。
To assess the effect of NtJAZ15 on tobacco glandular trichome development,the molecular mechanism underlying its involvement in trichome development was investigated.The NtJAZ15 gene was cloned from K326,and its bioinformatics analysis and tissue expression patterns were examined.By generating NtJAZ15 knockout and overexpression mutants,the regulatory roles of NtJAZ15 in glandular trichome formationwere analyzed.Results indicated that both NtJAZ15-1 and NtJAZ15-2 were highly conserved across different tobacco varieties,exhibiting typical TIFY and Jas domains characteristic of the JAZ family.NtJAZ15 was found to be expressed in all tissues,with NtJAZ15-2 exhibiting higher expression levels compared to NtJAZ15-1 in all tissues.Interestingly,the density of long and short glandular trichomes increased in NtJAZ15 knockout mutants,while the opposite effect was observed in overexpressed mutants.Furthermore,qRT-PCR analysis showed that NtJAZ15 significantly affected the expression of NtWo,NtCycB2,NtHD9 and NtHD12,but had no significant effect on the expression of NtMIXTA.These findings together suggest that NtJAZ15 negatively regulates the occurrence of long and short glandular trichome,and NtWo,NtCycB2,NtHD9 and NtHD12 may participate in the regulatory network involving NtJAZ15.
