4-苯基丁酸通过核转录因子-κB通路减轻滋养细胞内质网应激

4-phenylbutyric acid alleviates endoplasmic reticulum stress in trophoblasts through NF-κB pathway

摘要 目的探讨4-苯基丁酸(4-PBA)减轻内质网应激的信号通路。方法采用永生化的滋养细胞株HTR-8/Svneo为研究对象,建立内质网应激模型,加入4-苯基丁酸进行培养后,采用蛋白免疫印迹法比较质网应激特异性分子GRP78、蛋白激酶R样内质网激酶(PERK)、C/EBP同源蛋白(CHOP)的表达。同样的方法检测核转录因子(NF)-κB,caspase-3,B淋巴细胞瘤-2基因(Bcl-2)的表达。结果随着4-PBA浓度增加,内质网应激特征蛋白的表达量下降,各不同浓度之间表达量相比较,差异具有统计学意义。培养基中加入不同浓度的4-PBA进行培养后检测NF-κB的表达量随着4-PBA浓度的增加,NF-κB的表达量下降,差异具有统计学意义。结论4-苯基丁酸通过NF-κB通路减轻滋养细胞内质网应激。 Objective To explore the signaling pathway of 4-phenylbutyric acid(4-PBA)in alleviating endoplasmic reticulum stress.Methods A endoplasmic reticulum stress model was established using the immortalized trophoblast cell line HTR-8/Svneo.After adding 4-PBA for culture,the expression of endoplasmic reticulum stress specific molecules GRP78,PERK,and CHOP were compared using Western blot method.The same method was then used to detect the expression of NF-κB,caspase-3,and Bcl-2.Results As the concentration of 4-PBA increases,the expression level of endoplasmic reticulum stress characteristic proteins decreased,and the differences in expression levels between different concentrations were statistically significant.After culturing with different concentrations of 4-PBA added to the medium,the expression level of NF-κB decreased with the increase of 4-PBA concentration,and the difference was statistically significant.Conclusion 4-PBA alleviates endoplasmic reticulum stress in trophoblasts through the NF-κB pathway.
