崇明西红花花瓣中槲皮素含量测定及其应用探讨

Determination of Quercetin Content in Chongming Saffron Petals and Its Application Exploration

建立液相色谱联用质谱测定西红花花瓣中槲皮素的方法,精确测定花瓣中的槲皮素含量,以提升西红花非药用部分的综合利用提供参考。预试验表明,槲皮素易溶于部分有机溶剂,此试验中样品用70%乙醇作为提取溶剂,运用加温超声方式提取后,离心取上清液分析。采用ZORBAX Eclipse Plus C18色谱柱,以甲醇-5 mmol/L乙酸铵0.1%甲酸水溶液(75:25,V/V)为流动相,负离子模式,采用多反应监测(MRM)方式扫描,外标法定量。结果显示,槲皮素的检出限为5μg/kg,定量限为10μg/kg,槲皮素在300~2000μg/L范围内线性关系良好,线性关系R>0.999,精密度为1.2%,三水平的回收率分别为93.9%,95.9%和97.5%,三水平的精密度分别为0.90%,0.92%和3.31%。此研究前处理方法简单,提取溶剂低毒,分析准确度高,结果精密度好,可用于西红花中槲皮素的定量分析。

A method for determining quercetin in saffron petals by liquid chromatography-mass spectrometry is established to accurately determine the content of quercetin in the petals,providing reference for improving the comprehensive utilization of non-medicinal parts of saffron.Pre-experiments have shown that quercetin is easily soluble in some organic solvents.In this experiment,the sample is extracted using 70%ethanol water as the extraction solvent,and the supernatant is obtained by centrifugation after extraction using heated ultrasound.Using ZORBAX Eclipse Plus C18 chromatography column,methanol 5 mmol/L ammonium acetate 0.1%formic acid aqueous solution(75∶25,V/V)as mobile phase,negative ion mode,multiple reaction monitoring(MRM)scanning,and external standard quantification.The results show that the detection limit of quercetin is 5μg/kg,quantification limit of 10μg/kg,quercetin has good linearity within the range of 300 to 2000μg/L,with a linear relationship R>0.999 and a precision of 1.2%.The recovery rates at the three levels are 93.9%,95.9%,and 97.5%,respectively,and the precision at the three levels are 0.90%,0.92%,and 3.31%.This study has a simple pre-treatment method,low solvent toxicity,high analytical accuracy,and good precision,which can be used for the quantitative analysis of quercetin in saffron.
