微RNA-454-3p、FERM结构域蛋白6在乳腺癌组织中的表达水平及临床意义

摘要 目的探讨微RNA(miR)-454-3p、FERM结构域蛋白6(FRMD6)在乳腺癌组织中的表达水平以及临床意义。方法选取唐山市人民医院2016年12月至2018年12月住院手术的150例乳腺癌病人术中切除的乳腺癌组织标本及癌旁组织标本;采用实时荧光定量逆转录聚合酶链反应(qRT-PCR)检测miR-454-3p、FRMD6在组织中的水平;采用Kaplan-Meier法分析miR-454-3p、FRMD6水平与乳腺癌病人预后的关系,并用log-rank法进行检验;Cox回归分析影响乳腺癌病人预后的因素。结果乳腺癌组织miR-454-3p水平(1.25±0.24)高于癌旁组织(0.99±0.22),FRMD6水平(0.79±0.19)低于癌旁组织(1.01±0.24),差异有统计学意义(P<0.05);癌组织中miR-454-3p、FRMD6水平与淋巴结转移、组织分化以及TNM分期有关(P<0.05);miR-454-3p高表达组和FRMD6低表达组病人3年累积生存率分别低于miR-454-3p低表达组和FRMD6高表达组病人(均P<0.05);miR-454-3p高表达、FRMD6低表达、TNM分期为Ⅲ/Ⅳ期、淋巴结转移是影响乳腺癌病人预后的危险因素(P<0.05)。结论miR-454-3p在乳腺癌组织中表达上调,FRMD6在乳腺癌组织中表达降低,二者与乳腺癌病人预后密切相关,有望成为临床上预测乳腺癌病人预后的生物性标志物。 Objective To investigate the expression levels and clinical significance of miR-454-3p and FERM domain protein 6(FRMD6)in breast cancer tissues.Methods The breast cancer tissue specimens and adjacent tissue specimens were collected from 150 breast cancer patients who were hospitalized and operated in Tangshan people's Hospital from December 2016 to December 2018;realtime fluorescence quantitative PCR(qRT-PCR)was applied to detect the expression levels of miR-454-3p and FRMD6 in tissues;Kaplan-Meier method was applied to analyze the relationship between the levels of miR-454-3p and FRMD6 and the prognosis of breast cancer patients,the log-rank method was used to test;Cox regression was applied to analyze the factors affecting the prognosis of breast cancer patients.Pearson correlation was applied to analyze the miR-454-3p and FRMD6 levels in breast cancer tissues.Results The level of miR-454-3p in breast cancer tissue(1.25±0.24)was higher than that in adjacent tissue(0.99±0.22),and the level of FRMD6(0.79±0.19)was lower than that in adjacent tissue(1.01±0.24)(P<0.05);the levels of miR-454-3p and FRMD6 in breast cancer tissue were related to lymph node metastasis,tissue differentiation and TNM stage(P<0.05);the 3-year cumulative survival rate of patients with high miR-454-3p expression group and low FRMD6 expression group was obviously lower than that of patients with low miR-454-3p expression group and high FRMD6 expression group,respectively(P<0.05);high expression of miR-454-3p,low expression of FRMD6,TNM stageⅢ/Ⅳ,and lymph node metastasis were risk factors affecting the prognosis of breast cancer patients(P<0.05).Conclu⁃sions The expression of miR-454-3p is up-regulated in breast cancer tissue,and the expression of FRMD6 is down-regulated in breast cancer tissue.The two are closely related to the prognosis of breast cancer patients and are expected to be clinical biomarkers for predicting the prognosis of breast cancer patients.
