活性氧响应性纳米粒对小鼠氧化应激和炎症因子表达的影响

Effects of reactive oxygen species-responsive nanoparticles on oxidative stress and expressions of inflammatory factors in mice

摘要 目的探讨活性氧响应性纳米粒(OxbCD NP)对小鼠氧化应激和炎症因子表达的影响。方法20只小鼠随机均分为正常对照组、足趾肿胀模型组、OxbCD NP 1.0 mg/kg组和OxbCD NP 10 mg/kg组,观察OxbCD NP对小鼠足趾厚度的影响。20只小鼠随机均分为正常对照组、急性腹膜炎模型组、OxbCD NP 0.1 mg/kg组和OxbCD NP 1.0 mg/kg组,采用ELISA法检测小鼠腹腔灌洗液中过氧化氢(H_(2)O_(2))、TNF-α和IL-1β水平。18只小鼠随机均分为正常对照组、OxbCD NP 1000 mg/kg腹腔给药组和OxbCD NP 1000 mg/kg尾静脉给药组,观察大剂量OxbCD NP对小鼠血常规和肝肾功能的影响。结果与足趾肿胀模型组相比,OxbCD NP 10 mg/kg组足趾厚度减少(P<0.01)。与急性腹膜炎模型组相比,OxbCD NP 0.1 mg/kg组TNF-α和IL-1β水平降低(P<0.01),OxbCD NP 1.0 mg/kg组H_(2)O_(2)、TNF-α和IL-1β水平均降低(P<0.01)。与正常对照组相比,OxbCD NP 1000 mg/kg尾静脉给药组和OxbCD NP 1000 mg/kg腹腔给药组小鼠ALT、AST、血清尿素、SCr水平以及Hb、RBC、WBC、Plt均无统计学差异(P>0.05)。结论OxbCD NP具有良好的生物体内抗氧化和抗炎作用,并且生物安全性较高。 Objective To investigate the effects of reactive oxygen species-responsive nanoparticles(OxbCD NP)on the oxidative stress and expressions of inflammatory factors in mice.Methods Twenty mice were equally randomized into four groups of normal control group,toe swelling model group,OxbCD NP 1.0 mg/kg group and OxbCD NP 10 mg/kg group,and the effect of OxbCD NP on the toe thickness of mice was observed.Twenty mice were equally randomized into four groups of normal control group,acute peritonitis model group,OxbCD NP 0.1 mg/kg group and OxbCD NP 1.0 mg/kg group,and the levels of H_(2)O_(2),TNF-αand IL-1βin the peritoneal lavage fluid were detected by ELISA method.Eighteen mice were equally randomized into three groups of normal control group,intraperitoneal OxbCD NP 1000 mg/kg group and intravenous OxbCD NP 1000 mg/kg group,and the effects of large dose of OxbCD NP on the blood routine and hepatorenal function were observed.Results Compared with toe swelling model group,the toe thickness was decreased in OxbCD NP 10 mg/kg group(P<0.01).Compared with acute peritonitis model group,the levels of TNF-αand IL-1βwere decreased in OxbCD NP 0.1 mg/kg group(P<0.01),and the levels of H_(2)O_(2),TNF-αand IL-1βwere all decreased in OxbCD NP 1.0 mg/kg group(P<0.01).The levels of ALT,AST,serum urea,SCr,Hb,RBC,WBC and Plt in groups of intraperitoneal OxbCD NP 1000 mg/kg and intravenous OxbCD NP 1000 mg/kg were not different from those in normal control group(P>0.05).Conclusion OxbCD NP has good antioxidative and anti-inflammatory effects with higher biosafety in vivo.
