摘要
目的 探究心房颤动(简称房颤)患者左房心肌中胶原蛋白(Collagen)、弹性蛋白(Elastin)的表达水平以及调控弹性蛋白生成的基质金属蛋白酶(MMPs)、金属蛋白酶组织抑制剂(TIMPs)蛋白表达水平和纤维化程度。方法 收集房颤患者和窦性心律患者左房组织标本,用HE、Masson染色检测心肌纤维化,用免疫荧光法和蛋白免疫印迹法检测Collagen I、CollagenⅢ、Elastin、MMP-2、MMP-9和TIMP-2的蛋白表达变化。结果 与对照组(n=5)相比,房颤组(n=10)Collagen I、CollagenⅢ蛋白表达明显上调(0.42±0.17 vs 0.05±0.04,1.10±0.73 vs 0.23±0.15,P均<0.05),Elastin蛋白水平下降(0.47±0.27 vs 2.17±1.05,P<0.05);MMP-2、MMP-9蛋白表达增加,TIMP-2蛋白表达减少。结论 房颤左房弹性蛋白水平下降,胶原蛋白水平增加,引起纤维化程度加重,这一过程可能受MMP-2、MMP-9和TIMP-2的共同调控。
Objective To investigate the expression levels of collagen and elastin,matrix metalloproteinases(MMPs)and tissue inhibitors of metalloproteinases(TIMPs),and the degree of fibrosis in atrial fibrillation pa-tients.Methods Left atrial tissue samples from patients with atrial fibrillation and sinus rhythm were collected,myocardial fibrosis was detected by HE and Masson staining,and protein expression of Collagen I,Collagen III,E-lastin,MMP-2,MMP-9 and TIMP-2 were detected by immunofluorescence and western blotting.Results Com-pared with the control group,the protein expressions of Collagen I and Collagen II in AF group were significantly up-regulated,and the level of Elastin protein was decreased.The expression of MMP-2 and MMP-9 protein in-creased,while the expression of TIMP-2 protein decreased.Conclusion The decreased ratio of elastin/collagen in left atrium under atrial fibrillation leads to the aggravation of fibrosis,which may be co-regulated by MMP-2.MMP-9 and TIMP-2.
