人参不定根诱导、增殖及皂苷积累的研究

摘要 目的:建立人参Panax ginseng C.A.Mey.不定根诱导体系并优化液体培养增殖条件。方法:以MS为基础培养基,通过正交试验,研究生长素NAA和IBA对人参不定根诱导的影响;考察不同NAA、IBA浓度及其组合浓度对人参不定根生长和皂苷含量的影响。结果:诱导人参不定根最佳外植体类型为种胚,最佳固体培养基为MS+NAA 0.6 mg/L+IBA 1.0 mg/L+蔗糖3%+琼脂0.8%,其诱导率为88.4%,诱导时间为18 d。在不定根液体增殖培养中,IBA比NAA更有利于不定根的生长。分别添加NAA 2.0 mg/L、IBA 1.0 mg/L和NAA 1.0 mg/L+IBA 1.0 mg/L时,不定根生物量及皂苷产量均较高,其中以二者组合对人参不定根增殖效果最好,最佳培养基为MS+NAA 1.0 mg/L+IBA 1.0 mg/L+蔗糖3%。结论:建立了由人参不同外植体直接诱导不定根的方法,在短期内可获得大量不定根材料,为人参次生代谢产物的生产提供了新的药源。 Objective:To establish the induction system of Panax ginseng adventitious root and optimize the liquid culture and breeding conditions.Methods:The effects of auxin NAA and IBA on induction of Panax ginseng adventitious root were studied by or-thogonal experiment with MS as medium.The effects of different concentrations of NAA,IBA and their combinations on adventitious root growth and saponin content of Panax ginseng were investigated.Results:The seed embryo was the best type of explants to induce Panax ginseng adventitious root,and the best solid medium was MS with NAA 0.6 mg/L,IBA 1.0 mg/L,3%sucrose,0.8%agar,the induc-tion rate was 88.4%and induction time was 18 days.IBA was more beneficial to adventitious root growth than NAA in liquid culture.When NAA 2.0 mg/L,IBA 1.0 mg/L and NAA 1.0 mg/L with IBA 1.0 mg/L were added,the biomass and saponin content of adventi-tious root were higher,and the combination of NAA 1.0 mg/L and IBA 1.0 mg/L was the best for adventitious root proliferation.The optimal medium was MS with NAA 1.0 mg/L,IBA 1.0 mg/L,3%sucrose.Conclusion:The method of direct induction of adventitious root by different explants of Panax ginseng is established,and a large amount of adventitious root materials can be obtained in a short time,which provides a new drug source for the production of secondary metabolites of Panax ginseng.
