摘要
目的:研究鲜地黄Rehmannia glutinosa Libosch.中的酚酸类成分及其生物活性。方法:运用硅胶、MCI gel CHP-20、ODS、Sephadex LH-20葡聚糖凝胶等柱色谱方法和制备型高效液相对鲜地黄70%丙酮提取物进行系统的分离纯化,通过MS、NMR等手段,结合理化性质鉴定化合物结构。采用MTT法检测化合物对低氧诱导RPASMC细胞增殖的影响,并对所有化合物进行体外α-葡萄糖苷酶抑制活性筛选。结果:从鲜地黄中分离得到16个化合物,分别鉴定为:C-藜芦酰乙二醇(1)、桂皮酸(2)、阿魏酸(3)、顺式阿魏酸(4)、咖啡酸(5)、ω-hydroxypropioguaiacone(6)、对羟基苯乙醇(7)、毛蕊花糖苷(8)、贞桐苷A(9)、苯甲酸(10)、4-甲氧基苯甲酸(11)、对羟基苯甲酸(12)、香草酸(13)、香草醛(14)、3,4-二羟基苯甲醛(15)、丁香酸(16)。结论:其中,化合物1、4、6、9、11、15为首次从该属植物中分离得到。化合物1、15、16可显著抑制RPASMC细胞增殖且无明显毒性。所有化合物均显示微弱的降糖活性。
Objective:To study the phenolic acids from the fresh Rehmannia glutinosa and the biological activities.Methods:The compounds from the 70%acetone extract of fresh Rehmannia glutinosa were isolated and purified by silica gel,MCI gel CHP-20,ODS,Sephadex LH-20 and preparative HPLC.The structures were identified on the basis of MS,NMR and physicochemical properties.MTT method was used to evaluate the proliferation effect of the RPASMC cells and all isolated compounds were evaluated in vitro for theirα-glucosidase inhibitory activity.Results:Sixteen compounds were isolated from the fresh Rehmannia glutinosa and identified as C-veratroyl glycol(1),cinnamic acid(2),ferulaic acid(3),cis-ferulaic acid(4),caffeic acid(5),ω-hydroxypropioguaiacone(6),4-hydroxyphenethyl alcohol(7),acteoside(8),clerodenoside A(9),benzoic acid(10),anisic acid(11),4-hydroxybenzoic acid(12),vanillic acid(13),vanillin(14),3,4-dihydroxybenzene formaldehyde(15),syringic acid(16),respectively.Conclusion:Among them,compounds 1,4,6,9,11,and 15 are isolated from Rehmannia genus for the first time.Compounds 1,15,and 16 markedly inhibite the proliferation of RPASMC cells without obvious toxicity and all compounds show weakα-glucosidase inhibitory activity.
