量子点微球荧光免疫层析定量检测水产、畜禽肉组织中恩诺沙星

摘要 目的建立一种快速定量检测水产、畜禽肉组织中恩诺沙星的新方法学。方法以量子点荧光微球为新型标记探针,通过参数优化,制备获得量子点荧光微球免疫层析试纸条;进一步以T/C比值法建立定量检测恩诺沙星的新方法。结果在最优条件下,该试纸条检测缓冲液中恩诺沙星半数抑制浓度为1.04 ng/mL,检出限为0.13 ng/mL;检测水产、畜禽肉组织中恩诺沙星的定量检测范围为8.75~560.00μg/kg,半数抑制浓度为72.45μg/kg,加标回收实验显示该方法定量检测组织样本中恩诺沙星的回收率在83.03%~124.00%之间,变异系数小于15.00%,检测15个实际含有恩诺沙星的水产以及畜禽肉组织样本,其结果与液相色谱-串联质谱法检测结果高度一致;此外,加速保存实验结果显示该试纸条在室温下可保存一年。结论本研究以量子点荧光微球为新型标记探针,构建了高灵敏定量检测恩诺沙星的新方法,实现了水产、畜禽肉组织中恩诺沙星的简单、快速、定量分析。 Objective To establish a new methodology for rapid and quantitative detection of enrofloxacin in aquatic products and meat tissues of livestock and poultry.Methods Using quantum dot beads as a new labeling probe,the immunochromatographic test strip of quantum dot beads was prepared by parameter optimization.Further,a new method for quantitative detection of enrofloxacin was established by T/C ratio method.Results Under the optimum conditions,the half inhibitory concentration of enrofloxacin in the test buffer was 1.04 ng/mL,and the limit of detection was 0.13 ng/mL.The quantitative detection range of enrofloxacin in aquatic products and meat tissues of livestock and poultry was 8.75-560.00μg/kg,and the half inhibitory concentration was 72.45μg/kg.The recovery experiment showed that the recovery rates of enrofloxacin in tissue samples were 83.03%-124.00%,and the coefficient of variation was less than 15.00%.Fifteen aquatic products and meat tissues actually containing enrofloxacin were detected by this method.In addition,the accelerated storage experiment results showed that the test strip could be stored for one year at room temperature.Conclusion In this study,quantum dot beads aere used as new labeled probes,and a new method for quantitative detection of enrofloxacin with high sensitivity is constructed,which realizes the simple,rapid and quantitative analysis of enrofloxacin in aquatic products and livestock and poultry meat tissues.
