缢蛏Hsp基因家族全基因组鉴定及其在应激下的表达分析 被引量:1

Genome-wide Identification of Hsp Gene Family and Analyzing Its Expression Under Stress in Sinonvacula constricta

摘要 热休克蛋白(heat shock protein,Hsp)是一类广泛存在于动植物和微生物等多种生物体内的高度保守的蛋白质,能够被生物和非生物胁迫诱导表达。为了探究Hsp在缢蛏(Sinonovacula constricta)应对生物与非生物胁迫中的作用,本研究利用生物信息学方法,在缢蛏全基因组中共鉴定出62个ScHsp基因,对其基因结构、染色体定位和蛋白理化性质进行预测,并对ScHsp基因在不同组织和生物胁迫[副溶血弧菌(Vibrio parahemolyticus)感染]及非生物胁迫(高温和干露胁迫)下的表达模式进行分析。结果表明,不同ScHsp家族基因内含子的数目存在较大差异,62个ScHsp基因不均匀分布在17条染色体上,基因复制分析发现有3组串联复制基因,这可能是ScHsp家族基因数目扩张的重要原因;多数ScHsp基因表达具有组织特异性,少数基因(ScHsp90-2和ScHsp70-11)呈现泛表达模式;副溶血弧菌感染和高温胁迫后,ScHsp家族基因都呈现不同程度的响应,根据基因表达的趋势分别分为6种和4种模式;对低温和常温干露胁迫5个时间点(0、12、24、48、72 h)下,ScHsp基因家族6个代表基因的表达量进行qPCR分析,结果显示6个基因都不同程度地响应干露胁迫,且表达模式不同。本研究为全面深入研究ScHsp家族基因在缢蛏应对胁迫反应作用机制提供基础资料。 Heat shock proteins(Hsps),highly conserved proteins widely distributed in animals,plants and microorganisms,can be induced by biological and abiotic stress expression.In order to explore the role of Hsp in response to biotic and abiotic stresses in Sinonovacula constricta,this study identified 62 ScHsp genes in the whole genome of S.constricta by bioinformatics methods,and predicted their gene structure,chromosome location and protein physical and chemical properties,and the expression patterns of ScHsp family genes in S.constricta were analyzed in different tissues and under biological(Vibrio parahemolyticus infection)and abiotic stresses(high temperature stress and dry exposure).The results showed that there were significant differences in the number of introns in different ScHsp families;the 62 ScHsp genes unevenly distributed on 17 chromosomes,and gene replication analysis revealed three sets of tandem replication genes,which may be an important reason for the number expansion of ScHsp family genes;most of the ScHsp expressions were tissue-specific,while a few genes(ScHsp90-2 and ScHsp70-11)showed a pan-expression pattern;ScHsp family genes showed different levels of response to Vibrio parahaemolyticus infection and high temperature stress,and divided into 6 and 4 patterns according to the trend of gene expression;the expression levels of 6 representative genes of the ScHsp gene family were analyzed by qPCR at 5 time points(0,12,24,48,72 h)under low temperature and normal temperature dry exposure.The results showed that the 6 genes responded to dry exposure to different degrees.This study provides basic information for a comprehensive and in-depth study of the mechanism of ScHsp genes in response to stress in S.constricta.
