摘要
研究五味子水提物对非酒精性脂肪肝(nonalcoholic fatty liver disease,NAFLD)小鼠肠道菌群的影响。采用高脂饮食饲喂16周诱导NAFLD小鼠模型,从第4周五味子水提物灌胃给药;16周后检测血清ALT、AST、TC和TG水平;HE和油红O染色观察肝脏病理变化;免疫荧光染色观察肝脏IL-6和TNF-α表达;高通量测序技术分析结肠内容物肠道菌群变化。结果表明,与模型组比较,五味子水提物高剂量组体质量明显下降;低剂量和高剂量组肝脏质量明显下降;各给药组ALT、AST、TC和TG水平明显降低;肝脏病理和炎症均明显改善;肠道菌群多样性及上述优势物种丰度均明显纠正,且菌群的改变与药效学指标具有相关性。五味子水提物对NAFLD小鼠表现出确切的治疗作用,且与改善其肠道菌群相关。
To study the effects of Schisandra chinensis aqueous extract on intestinal flora in nonalcoholic fatty liver disease(NAFLD)mice,a NAFLD model was established by high-fat diet feeding mice for 16 weeks,and Schisandra chinensis aqueous extract was administered by gavage from week 4.Serum ALT,AST,TC and TG levels were measured after 16 weeks.HE and oil red O staining were used to observe pathological changes of mice liver.Immunofluorescence staining was used to observe expressions of IL-6 and TNF-αin mice liver.High-throughput sequencing was used to analyze the changes of intestinalflora in colonic contents.The results indicated that,compared with the control group,the body mass in the mice group with high-dose Schisandra chinensis aqueous extract was significantly reduced.The liver mass in mice groups with low-dose and high-dose Schisandra chinensis aqueous extract was significantly reduced;ALT,AST,TC and TG in each administration group were significantly reduced;mice liver pathology and inflammation were significantly alleviated;the diversity of intestinalflora and abundance of above-mentioned dominant species were significantly corrected,and changes of flora were correlated with pharmacodynamic indexes.Schisandra chinensis aqueous extract had a definite therapeutic effect on NAFLD mice,which might be associated with its improvement of intestinalflora in NAFLD mice.
