HPLC法同时测定三味拳参口服液中苦参碱和氧化苦参碱的含量

摘要 试验旨在建立利用高效液相色谱法(HPLC)同时测定三味拳参口服液中苦参碱和氧化苦参碱的含量。试验对比了3种氨基色谱柱(安捷伦ZORBAX NH_(2)、资生堂CAPCELL PAK NH_(2)、Waters SPHERSORB NH_(2))的检测效果。从分离效果角度考虑,优先使用安捷伦ZORBAX NH_(2),但其运行时间长;从节约时间和流动相等试剂角度考虑,推荐优先使用资生堂CAPCELL PAK NH_(2)色谱柱,次选Waters SPHERSORB NH_(2)色谱柱。三味拳参口服液经无水乙醇提取后,采用氨基色谱柱(4.6 mm×250 mm,5μm)进行分离,柱温35℃,检测波长220 nm。采用二极管阵列检测器进行分析,外标法进行定量测定。结果表明,在30~300 mg/L的范围内,苦参碱和氧化苦参碱线性关系良好(R>0.999 0),平均加样回收率为97.64%,变异系数为1.63%。系统适应性试验和色谱柱耐受性良好,完全能够满足检测的需求。研究表明,试验建立的方法具有操作简便、重复性好、稳定性高、准确性强等优点,可以用于三味拳参口服液中苦参碱和氧化苦参碱含量的准确检测。 The study was to establish a method for the simultaneous determination of matrine and oxymatrine in Sanwei Quanshen Koufuye by HPLC.The test compares the detection effects of three amino-chromatographic columns(Agilent ZORBAX NH_(2),SHISEIDO CAPCELL PAK NH_(2),Waters SPHERSORB NH2).For separation effect,Agilent ZORBAX NH_(2) is preferred,but its running time is long.In order to save time and flow equivalent reagents,it was recommended to use SHISEIDO CAPCELL PAK NH_(2) column first and subselect Waters SPHERSORB NH_(2) column.After the absolute ethanol extraction,the amino chromatography column(4.6 mm×250 mm,5μm)was used as the separation column,column temperature was 35℃,and detection wavelength was 220 nm.Diode array detector was used for analysis,and external standard method was used for quantitative determination.The results showed that matrine and oxymatrine showed a good linear relationship ranging between 30 and 300 mg/L(R>0.9990),and the average sample recovery rate was 97.64%with RSD 1.63%.The system adaptation test and the chromatography column tolerance was good,which fully meet the detection requirements.The study indicates that the method is easy to operate,with good repeatability,stability and accuracy,and can be used for the detection of matrine and oxymatrine contents in Sanwei Quanshen Koufuye.
