摘要
目的:对2型糖尿病大鼠勃起功能障碍(ED)的发生与FGF21的相关性进行研究,观察FGF21与糖尿病并发勃起功能障碍(DMED)发生的关系,分析FGF21在DMED发生中的作用。方法:雄性SPF级SD大鼠40只,大随机分为正常对照组(n=10)和糖尿病组(n=30),糖尿病组大鼠给予注射链脲佐菌素(STZ)构建糖尿病模型。皮下注射阿扑吗啡(APO)溶液,诱导阴茎勃起,记录大鼠阴茎勃起频率,根据阴茎有无勃起和勃起的次数再分为勃起障碍组(DMED组)和无勃起障碍组(DM组)。取三组大鼠血清测定大鼠血清FGF21水平;观察期后,分离大鼠阴茎组织,进行Masson染色观察三组大鼠阴茎组织的病理变化。结果:最终DMED组大鼠有12只,DM组13只,正常对照组13只。对照组、DM组及DMED组三组大鼠的FGF21浓度分别为(114.9±5.6)pg/mL、(160.32±7.2)pg/mL、(241.1±8.5)pg/mL,DMED组FGF21水平明显升高。DMED组大鼠阴茎组织平滑肌大量增生;与对照组相比,海绵体窦腔大量减少,窦腔甚至消失,周围结缔组织长入。结论:FGF21在糖尿病大鼠体内高表达,与DMED发生密切相关,是糖尿病性勃起功能障碍一个新的提示因素及危险因素。
Objective:To study the correlation between the occurrence of erectile dysfunction(ED)and FGF21 in type 2 diabetes rats,observe the relationship between FGF21 and the occurrence of diabetetes mellitus induced erectile dysfunction(DMED),and analyze the role of FGF21 in the occurrence of DMED.Methods:40 male SPF grade SD rats were randomly divided into normal control group(n=10)and diabetes group(n=30).The rats in diabetes group were injected with streptozotocin(STZ)to establish diabetes model.Subcutaneously inject apomorphine(APO)solution to induce penile erection,record the frequency of penile erection in rats,and divide them into erectile dysfunction group(DMED group)and non erectile dysfunction group(DM group)based on the presence and frequency of penile erection.Take three groups of rat serum to measure the level of FGF21 in rat serum.After the observation period,the penile tissue of rats was separated and Masson staining was used to observe the pathological changes of the penile tissue of the three groups of rats.Results:12 rats in the DMED group,13 rats in the DM group,and 13 rats in the normal control group.The concentrations of FGF21 in control group,DM group and DMED group were(114.9±5.6)pg/mL,(160.32±7.2)pg/mL and(241.1±8.5)pg/mL,respectively.The levels of FGF21 in DMED group were significantly higher.In DMED group,the smooth muscle of penis tissue proliferated significantly.Compared with the control group,the cavernous sinus cavity was significantly reduced,and even disappeared,with surrounding connective tissue growing in.Conclusion:High expression of FGF21 in diabetic rats,and closely related to the occurrence of DMED,which is a new suggestive factor and risk factor of erectile dysfunction in diabetes.
