摘要
天然产物是创新药物先导化合物的重要来源,具有结构多样性以及功能独特性。目前,天然产物在癌症、心脑血管疾病、神经系统疾病、自身免疫性疾病等的治疗中发挥重要作用。同时,蛋白质靶向嵌合体(PROTAC)技术的兴起为临床分子靶向药物耐药提供了新的解决方案,天然产物及其衍生物作为PROTAC的重要组成部分,在靶向蛋白降解药物的开发中发挥着重要作用。综述了基于天然产物的PROTAC以及PROTAC技术在天然产物靶标鉴定中的应用进展。
Natural products are the important sources of lead compounds for innovative drugs,exhibiting structural diversity and functional specificity.At present,natural products play an important role in the targeted therapy of cancer,cardiovascular and cerebrovascular diseases,nervous system diseases,and autoimmune diseases.The development of protein degradation targeted chimeras(PROTAC)provides a new strategy for clinical molecular targeted drug resistance.Natural products and their derivatives,as important parts of PROTAC,play an important role in the development of protein targeted drugs.This article reviewed the progress in the application of natural product-based PROTAC,as well as PROTAC technology for the identification of natural products targets.
