摘要
目的探讨微小RNA-506(miR-506)对急性胰腺炎大鼠(AP)巨噬细胞迁移抑制因子(MIF)及肠屏障功能的影响。方法将SPF级SD大鼠50只随机均分为对照组、模型组、miR-506组、MIF组、miR-506+MIF组。观察各组胰腺组织病理学变化、观察胰腺组织病理学变化、肠屏障功能[血清淀粉酶、内毒素、血浆D-乳酸、乳果糖/甘露醇排除率(L/M)、细菌移位率]及MIF水平变化。。结果HE染色显示,对照组胰腺组织形态结构正常,无明显的病理改变;模型组、MIF组、miR-506组及miR-506+MIF组均有不同程度胰腺坏死,MIF组病变最为严重;miR-506组病变最轻。miR-506组胰腺病理评分、血清淀粉酶、内毒素、MIF、血浆D-乳酸水平、L/M、细菌移位率及MIF mRNA相对表达量较其他干预组降低,miR-506相对表达量则升高(P<0.05);miR-506+MIF组血清淀粉酶、内毒素、MIF、血浆D-乳酸水平、L/M、细菌移位率及MIF mRNA相对表达量较miR-506组升高,miR-506相对表达量较miR-506组下降(P<0.05)。结论miR-506的高表达减轻AP大鼠病变,改善肠屏障功能,其作用可能与抑制MIF有关。
Aim To investigate the effects of microRNA-506(miR-506)on macrophage migration inhibitory factor(MIF)and intestinal barrier function in acute pancreatitis(AP)rats.Methods Fifty SPF grade SD rats were selected to establish AP model and treated with miR-506 and MIF.Pancreatic histopathological changes,intestinal barrier function[serum amylase,endotoxin,plasma D-lactic acid,lactulose/mannitol exclusion rate(L/M),bacterial translocation rate]and MIF level were observed.Results HE staining showed that the control group had normal pancreatic tissue morphology and structure,with no obvious pathological changes.Model group,MIF group,miR-506 group and miR-506+MIF group all had different degrees of pancreatic necrosis,and the lesions in MIF group were the most serious.miR-506 group had the mildest pathological changes.The pancreatic pathological score,serum amylase,endotoxin,MIF,plasma D-mammary gland level,lactulose/mannitol exclusion rate(L/M),bacterial translocation rate and MIF mRNA relative expression level of miR-506 group were lower than those of other groups,while the relative expression level of miR-506 was increased(P<0.05).After dual intervention with miR-506 and MIF,serum amylase,endotoxin,MIF,plasma D-mammary gland level,L/M,bacterial migration rate and relative expression level of MIF mRNA in miR-506+MIF group were higher than those in miR-506 group,and the relative expression level of miR-506 was lower than that in miR-506 group(P<0.05).Conclusion The high expression of miR-506 alleviates the lesions of AP rats and improves the intestinal barrier function,which may be related to the inhibition of MIF.
