复方三黄凝胶薄层色谱鉴别方法研究

摘要 目的建立复方三黄凝胶的薄层色谱(TLC)鉴别方法。方法采用TLC考察不同展开剂、不同温度、不同薄层板厂家对复方三黄凝胶中大黄、黄芩、黄柏鉴别的影响,确定最佳TLC条件。结果大黄采用硅胶H薄层板,以甲苯-醋酸乙酯-甲酸(7∶3∶0.1)为展开剂,取出,晾干,置365 nm紫外光灯下检视,置氨蒸气中熏后,斑点显色清晰;黄芩采用聚酰胺薄膜,以甲苯-醋酸乙酯-甲醇-甲酸(10∶3∶1∶2)为展开剂,展开,取出,晾干,置365 nm紫外光灯下检视,斑点显色清晰;黄柏采用硅胶G薄层板,以甲苯-甲醇-醋酸乙酯-异丙醇-浓氨试液(6∶1.5∶3∶1.5∶0.5)为展开剂,展开,取出,晾干,置365 nm紫外光灯下检视,斑点显色清晰,三味药的TLC图特征明显,分离度好且无阴性干扰。结论所用TLC方法专属性强,耐用性好,可作为复方三黄凝胶的质量控制方法。 Objective To establish the thin layer chromatography(TLC)method for identification of compound Sanhuang Gel.Methods The TLC was used to investigate the influence of different developing agent,different temperature and different thin layer plate manufacturers on the identification of rhubarb,scutellaria and phellodendron in compound Sanhuang Gel,and to determine the best TLC conditions.Results The rhubarb was made of silica gel H thin layer plate with toluene-ethyl acetate-formic acid(7∶3∶0.1)as the development agent,removed,dried,examined under 365 nm UV lamp,the spots showed clear color.Polyamide film was used in scutellaria baicalensis,and toluene-ethyl acethane-methanol-formic acid(10∶3∶1∶2)was used as development agent,removed,dried,examined under 365 nm UV lamp,and fumed in ammonia vapor,the spots showed clear color.The silica gel G thin layer plate was used and toluene-methanol-ethyl-acetate-isopropyl-alcohol-concentrated ammonia test solution(6∶1.5∶3∶1.5∶0.5)was used as the development agent,removel,dried,and examined under 365 nm ultraviolet lamp,the spots showed clear color.The TLC of the three drugs showed obvious characteristics,good separation and no negative interference.Conclusion The TCL method is specific and durable,which can be used for the quality control of compound Sanhuang Gel.
