鲁丽苹果叶片高效不定芽再生体系的优化

Optimization of high-efficiency adventitious bud regeneration system from leaf explants of Malus domestica var.‘Luli’

摘要 以鲁丽和GL-3等苹果组培苗离体叶片为外植体,研究了极性及伤口处理对叶片高效不定芽再生的影响。结果表明:放置叶片时,远轴面接触培养基时,鲁丽和GL-3叶片不定芽再生率分别为81.14%、93.21%;近轴面接触培养基时,叶片不定芽再生率分别为92.71%和95.73%,且鲁丽叶片平均不定芽数高于GL-3。处理伤口时,鲁丽整叶横切叶片不定芽再生率为91.20%,显著高于0.8 cm×0.5 cm横切处理;GL-3整叶横切叶片不定芽再生率也高于0.8 cm×0.5 cm横切处理,且2个品种叶片横切处理后不定芽玻璃化程度均显著降低。鲁丽不定芽再生,最适叶片放置方式为近轴面接触培养基,适宜伤口处理方式为整叶横切,研究结果为种质资源的挖掘利用和基因功能验证奠定基础。 The influence of polarity and wound treatment on efficient adventitious bud regeneration from excised leaves of the apple cultivars‘Luli’and‘GL-3’was studied.The results showed that when the leaves were placed with the abaxial surface in contact with the culture medium,the regeneration rates of‘Luli’and‘GL-3’leaves were 81.14%and 93.21%,respectively.When the proximal surface was in contact with the culture medium,the regeneration rates were 92.71%and 95.73%,respectively,with‘Luli’exhibiting a higher average number of adventitious buds compared to‘GL-3’.Regarding wound treatment,the regeneration rate of‘Luli’leaves subjected to whole leaf transsection was 91.20%,significantly higher than that of 0.8 cm×0.5 cm transsection treatment.Similarly,the regeneration rate of‘GL-3’leaves subjected to whole leaf transsection was higher than that of 0.8 cm×0.5 cm transsection treatment,and the degree of vitrification of adventitious buds significantly decreased after transsection treatment in both varieties.For adventitious bud regeneration in‘Luli’,the most suitable leaf placement was with the proximal surface in contact with the culture medium,and the appropriate wound treatment method was whole leaf transsection.This study lays the foundation for the exploration and utilization of germplasm resources and the validation of gene functions.
