一种含1,2,3-三氮唑席夫碱的Al^(3+)荧光探针及其应用

摘要 本文设计合成了5-苯基-2-苯基-1,2,3-三唑-1-4-酰肼(PP),将其与水杨醛反应,构建了一种席夫碱荧光探针(PPS)。荧光光谱及紫外光谱测定表明,PPS能够高选择性和强抗干扰性地检测乙醇溶液中的Al^(3+),检出限为8.28 nmol/L;在紫外光下可发生明显的溶剂变色效应,在含水量大于60%的有机溶剂中存在聚集诱导发光(AIE)效应,质谱、工作曲线、核磁滴定及理论计算结果均证明,PPS和Al^(3+)可以络合比1∶1进行反应,生成平面结构的化合物。该探针具有低的细胞毒性且成功用于HeLa细胞的显色成像,有望被开发利用为具有AIE聚集效应的发光材料及生物体内测定Al^(3+)的荧光探针。 5-phenyl-2-phenyl-1,2,3-triazole-4-hydrazid(PP)was designed and synthesized based on"clickchemistry".Thefluorescenceprobe PPS(5-phenyl-2-phenyl-1,2,3-triazole-4-hydrazid-0-hydroxymethylbenzene)containing Schiff base was further constructed by a chemical reaction between PP and salicylaldehyde.PPS could be used to determine Al^(3+)in ethanol solution with high sensitivity and strong antiinterference performance by fluorescence and UV-vis spectroscopic methods.The limit of detection for Al^(3+)was 8.28 nmol/L,and the probe PPS showed an obvious solvatochromic effect under ultraviolet light.The aggregationinduced emission(AIE)effect of PPS was found in organic solvent when water content was more than 60%.Furthermore,the results of mass spectrometry(MS),Job's plot,NMR titration,and density functional theory(DFT)calculation confirmed that the probe PPS and Al^(3+)could react to form a compound with a planar structure at 1:1 complexation ratio.The probe PPS showed less low cytotoxicity to HeLa cells,so it was successfully used for chromatic bioimaging Al^(3+)in HeLa cells.It is expected to be developed and utilized as a kind of luminous material with AIE effect and a fluorescence probe for detecting A13*in vivo.
