常用酰基辅酶A的合成

摘要 酰基辅酶A作为重要的酰基活化形式在代谢中起着重要作用,而获得酰基辅酶A是进行相关代谢过程研究的基础。目前酰基辅酶A的合成方法主要为化学合成和酶法合成两类,不同结构的酰基辅酶A适用的合成方法有所不同。本文采用化学合成(酸酐合成、咪唑基活化和PyBOP一步合成)和酶法合成(辅酶A连接酶和酰基转移酶完成体外催化),合成了苯乳酰辅酶A、异戊酰辅酶A、苯甲酰辅酶A、对香豆酰辅酶A和肉桂酰辅酶A,并对合成结果进行了评价。结果表明,酶法具有转化率高,专一性强的优势,化学合成具有高效快速合成的特点。本文为将来合理选择酰基辅酶的合成方法提供了参考。 Acyl-coenzyme A,as an important form of acyl activation,plays a crucial role in metabolism,and obtaining acyl-coenzyme A is the basis for studying related metabolic processes.Currently,the synthesis methods of acyl-coenzyme A mainly include chemical synthesis and enzymatic synthesis,and the applicable synthesis methods vary for different structures of acyl-coenzyme A.In this paper,chemical synthesis,namely anhydride synthesis,imidazole activation,and PyBOP one-step synthesis,and enzymatic synthesis,namely CoA ligase and acyl transferase complete in vitro catalysis are used to synthesize phenyllactyl coenzyme A,isovaleryl coenzyme A,benzoyl coenzyme A,coumaroyl coenzyme A,cinnamoyl coenzyme A respectively,and the synthesis results are evaluated.The results show that enzymatic method has the advantages of high conversion rate and high specificity,and chemical synthesis has the characteristics of efficient and rapid synthesis.This paper provides a reference for the rational selection of acyl-coenzyme synthesis methods in the future.
