摘要
为建立鸡传染性法氏囊病病毒ELISA抗体检测试剂盒,该研究以灭活纯化的鸡传染性法氏囊病病毒作为包被抗原,连续制备了3批次鸡传染性法氏囊病病毒ELISA抗体检测试剂盒中试产品。结果表明:该试剂盒对IBDV阳性血清检测的灵敏度可达1∶6400;对鸡新城疫病毒、鸡减蛋下降综合征病毒、鸡传染性支气管炎病毒、鸡抗网状内皮组织增生症病毒、鸡马立克氏病病毒、禽流感病毒H5亚型、鸡传染性贫血病毒阳性血清和SPF鸡阴性血清均无交叉反应;批内和批间重复性的变异系数分别在4.21%和5.07%以内。应用该试剂盒和某进口同类ELISA试剂盒对258份临床白羽肉鸡血清样品同时进行对比检测,相对敏感性、相对特异性和符合率分别为98.44%、96.97%和98.06%。可见,该试验建立的鸡传染性法氏囊病病毒ELISA抗体检测试剂盒具有有操作简便、特异性强、灵敏度高的特点。
In order to develop an indirect enzyme-linked immunosorbent assay(ELISA)kit for the detection of antibodies against Infectious bursaldisease virus(IBDV),3 batches of IBDV ELISA antibody detection kits were prepared with inactivated and purified IBDV as coated antigen.The results showed that the sensitivity of the kit to IBDV positive serum was 1∶6400;There were no cross-reaction to the anti-sera against Newcastl Disease Virus,Egg Drop Syndrome Virus,Infectious Bronchitis Virus,Reticuloendotheliosis Virus,Marek's Disease Virus,Avian Influenza Virus H5 subtype,Chicken Infectious Anemia Virus and negative serum of SPF;The coefficient of variation of intra-lot and inter-lot repeatability was within 4.21%and 5.07%,respectively.A total of 258 serum samples of clinical white feather broilers were tested by this kit and an imported ELISA kit,the relative sensitivity,relative specificity and coincidence rates were 98.44%,96.97%and 98.06%,respectively.It can be seen that the ELISA antibody detection kit for avian infectious bursal disease virus established in this study has the characteristics of simple operation,strong specificity and high sensitivity.
