摘要
目的:观察玉屏风加味鼻喷剂对变应性鼻炎大鼠鼻腔水液代谢的作用,探讨其改善变应性鼻炎大鼠流涕症状的相关机制。方法:采用卵清蛋白(OVA)与氢氧化铝[Al(OH)3]联合致敏建立变应性鼻炎大鼠模型,随机分为模型组、玉屏风加味鼻喷剂组(YPF+,50μg/侧滴鼻,每日2次)及阳性对照组(糠酸莫米松,50μg/侧滴鼻,每日1次),另设空白组。空白组和模型组给予等量生理盐水(50μg/侧滴鼻,每日2次),连续4周。对比各组大鼠行为学评分、鼻黏膜HE染色病理并量化评分、PAS染色后计数杯状细胞、蛋白免疫印迹及免疫荧光法检测水通道蛋白5(AQP5)的表达情况。结果:与空白组相比,模型组大鼠行为学总评分及流涕症状评分均显著升高(P<0.05),鼻黏膜腺体增生及黏膜下层水肿明显,病理评分及杯状细胞计数显著升高(P<0.05),AQP5荧光表达明显减弱,蛋白水平显著降低(P<0.05);与模型组相比,YPF+组大鼠行为学评分及流涕症状评分均显著降低(P<0.05),鼻黏膜结构修复、炎性浸润改善、腺体增生好转,病理评分下降及杯状细胞计数显著减少(P<0.05),鼻黏膜AQP5蛋白荧光标记重现,蛋白水平显著回升(P<0.05)。结论:YPF+可缓解大鼠流涕,改善变应性鼻炎大鼠鼻腔水液代谢,其机制可能与抑制变应性鼻炎大鼠鼻黏膜杯状细胞增生及上调AQP5的表达相关。
Objective The study was aimed at observing the effect of Modified Yupingfeng nasal spray(YPF+)on nasal fluid metabolism in rats with allergic rhinitis(AR)and exploring the related mechanism of YPF+to improve the symptoms of runny nose in AR rats.Methods The AR rat model was sensitized by Ovalbumin(OVA)and aluminum hydroxide[Al(OH)_(3)],which was randomly divided into model group,Yupingpeng nasal spray group(YPF+,50μg/side nasal drip,twice a day),positive control group(Mometasone furoate,50μg/side nasal drip,once a day),and blank group.The blank group and the model group were given the same amount of normal saline(50μg/side nasal drops,twice a day)for 4 weeks.The behavioral scores,pathological and quantitative scores of HE staining in nasal mucosa,goblet cell counting after PAS staining,and aquaporin 5(AQP5)expression detected by Western blot and immunofluorescence were compared in each group.Results Compared with the blank group,the total behavioral score and runny nose symptom score of rats in model group were significantly increased(P<0.05).Meanwhile,the mucosal glandular hyperplasia and submucosal edema,the pathological score and goblet cell counts were significantly increased(P<0.05).On the contrary,the fluorescence expressionand the protein level of AQP5 was significantly decreased,(P<0.05).Compared with the model group,total behavioral score and single symptom score of runny nose in YPF+group were significantly decreased(P<0.05).Structural repair,inflammatory infiltration and glandular hyperplasia of nasal mucosa were improved.Pathological score and goblet cell counts were decreased significantly(P<0.05).Meanwhile,the fluorescence labeling of AQP5 protein appeared in nasal mucosa,and the protein level increased significantly(P<0.05).Conclusion YPF+can relieve runny nose and improve nasal fluid metabolism in AR rats.Its mechanism may be connectedwith inhibiting goblet cell proliferation and up-regulating AQP5 expression in AR rats nasal mucosa.
