摘要
采用基于免疫磁珠净化的高效液相色谱技术测定植物油中黄曲霉毒素B_(1)(AFB_(1))、黄曲霉毒素B_(2)(AFB_(2))、黄曲霉毒素G1(AFG1)和黄曲霉毒素G_(2)(AFG_(2))含量。样品以甲醇-水(70∶30,体积比)提取后,经黄曲霉毒素总量免疫磁珠试剂盒净化,并结合免疫磁珠自动工作站,显著提升了净化效率。4种黄曲霉毒素在一定质量浓度范围内具有良好的线性关系,相关系数(r^(2))均大于0.99,AFB_(1)与AFB_(2)、AFG1与AFG_(2)的定量下限分别为0.05μg/kg和0.10μg/kg。4种毒素在5种商品植物油基质中的平均回收率为74.0%~96.0%,日间和日内相对标准偏差(RSD)均小于7.0%。所建立的方法与国标GB 5009.22-2016的免疫亲和柱法无显著性差异,方法准确、灵敏,适用于植物油中4种黄曲霉毒素的同时定量分析。
In this study,a high performance liquid chromatography method based on immunomag⁃netic beads purification for the determination of aflatoxin B_(1)(AFB_(1)),aflatoxin B_(2)(AFB_(2)),aflatoxin G1(AFG1)and aflatoxin G_(2)(AFG_(2)))in vegetable oils was established.First,the aflatoxins in the sample were extracted with methanol-water(70∶30,volume ratio).The samples were purified with aflatoxin total immunomagnetic bead kit,combined with the immunomagnetic bead automatic work⁃station,which significantly improved the purification efficiency.Aflatoxins showed good linearity with correlation coefficients(r^(2))more than 0.99.The limits of quantitation(LOQs)for AFB_(1)and AFB_(2),AFG1 and AFG_(2) were 0.05μg/kg and 0.10μg/kg,respectively.The average recoveries of af⁃latoxins in five commercial vegetable oils ranged from 74.0%to 96.0%,with the intra-day and intraday relative standard deviations(RSDs)less than 7.0%.The results of the comparison experiments showed that the method had the same performance as the national standard GB 5009.22-2016.The established method has good accuracy and sensitivity,which is suitable for simultaneous quantitative detection of four aflatoxins in vegetable oils.
