长链非编码RNA基因多态性与原发性肝癌遗传易感性的相关性研究

摘要 目的探讨长链非编码RNA(lncRNA)基因单核苷酸多态性(SNP)与原发性肝癌遗传易感性的相关性,为原发性肝癌的早诊断、早预防、开展肝癌遗传易感机制研究提供依据。方法选取41例原发性肝癌患者和83例健康志愿者分别作为病例组和对照组,对浆细胞瘤变体易位基因1(PVT1)、结肠癌相关转录本1(CCAT1)、结肠癌相关转录本2(CCAT2)、肿瘤易感性8(CASC8)、前列腺癌相关非编码RNA 1(PRNCR1)中的13个SNP位点进行基因分型,比较两组患者的基因型分布。结果病例组与对照组lncRNA PVT1基因RS2392885位点的基因型分布比较,差异有统计学意义(P﹤0.05),其他位点基因型分布比较,差异均无统计学意义(P﹥0.05)。lncRNA PVT1基因上RS2392885位点的基因型为CC、CT和TT型,其中病例组各基因型占比分别为12.2%、68.3%和19.5%,对照组各基因型占比分别为19.3%、42.2%和38.6%。结论lncRNA PVT1中的RS2392885位点的基因型可能与原发性肝癌的遗传易感性有关。 Objective To investigate the correlation between gene single nucleotide polymorphisms(SNP)of long non-coding RNA(lncRNA)and genetic susceptibility of primary liver cancer,and to provide basis for the early diagnosis,prevention and the study of genetic susceptibility mechanism of liver cancer.Method A total of 41 patients with primary liver cancer and 83 healthy volunteers were selected as the case group and the control group,respectively.Plasmacytoma variant translocation 1(PVT1),colon cancer associated transcript 1(CCAT1),colon cancer associated transcript 2(CCAT2),cancer susceptibility 8(CASC8),prostate cancer associated non-coding RNA 1(PRNCR1)were genotyped for 13 SNP loci.The genotype distribution was compared between the two groups.Result The difference was statistically significant in the genotype distribution of RS2392885 locus of lncRNA PVT1 gene between the case group and the control group(P<0.05),and there were no significant differences in the genotype distribution of other loci(P>0.05).The genotypes of RS2392885 locus in lncRNA PVT1 gene were CC,CT and TT types,and the proportions of each genotype in the case group were 12.2%,68.3%and 19.5%,respectively,and those in the control group were 19.3%,42.2%and 38.6%,respectively.Conclusion The genotype of RS2392885 locus in lncRNA PVT1 may be associated with the genetic susceptibility of primary liver cancer.
