不同发酵工艺对浙产六神曲中5种成分含量变化的影响

摘要 目的建立同时测定浙产六神曲中苦杏仁苷、香草酸、芦丁、阿魏酸、槲皮素的高效液相色谱法,并考察不同发酵条件对浙产六神曲中5个成分含量变化的影响。方法对同一批次六神曲样品,通过设置不同的发酵条件(温度为25℃、30℃、35℃,相对湿度为60%、70%、80%、90%)进行发酵并分别取样。色谱柱为Chrom Core AR C18柱(4.6 mm×250 mm,5μm);以乙腈为流动相A,0.08%磷酸水溶液为流动相B,进行梯度洗脱;流速为1.0 ml/min,柱温为30℃,检测波长220 nm。分析苦杏仁苷、香草酸、芦丁、阿魏酸、槲皮素5种成分的变化。结果苦杏仁苷、香草酸、芦丁、阿魏酸、槲皮素在选定质量浓度范围内线性关系、重复性、稳定性良好,平均加样回收率均达到要求。浙产六神曲最佳发酵工艺参数为温度30℃,相对湿度80%,发酵时间为3 d。结论本研究建立的方法操作简便、结果准确、重复性及稳定性均较好,可用于浙产六神曲的质量控制。浙产六神曲最佳发酵工艺参数为温度30℃,相对湿度80%,发酵时间为3 d。 Objective To establish a high performance liquid chromatography for simultaneous determination of amygdalin,vanillic acid,rutin,ferulic acid,and quercetin in Massa Medicata Fermentata produced in Zhejiang Province,and to investigate the effects of different fermentation conditions on contents changes of five components in Massa Medicata Fermentata produced in Zhejiang Province.Methods The same batch of Massa Medicata Fermentata sample was fermented by setting different fermentation conditions(temperature was 25℃,30℃,and 35℃,relative humidity was 60%,70%,80%,and 90%)and sampled respectively.The chromatography was performed on ChromCore AR C18 column(4.6 mm×250 mm,5μm);acetonitrile as mobile phase A and 0.08%phosphoric acid aqueous solution as mobile phase B were used for gradient elution;the flow rate was 1.0 ml/min,the column temperature was 30℃,and the detection wavelength was 220 nm.The changes of five components of amygdalin,vanillic acid,rutin,ferulic acid,and quercetin were analyzed.Results The linearity,repeatability,and stability of amygdalin,vanillic acid,rutin,ferulic acid,and quercetin in the selected concentration range were good,and the average recovery rate reached the requirement.The optimum fermentation parameters of Massa Medicata Fermentata produced in Zhejiang Province were 30℃of temperature,80%of relative humidity,and 3 d of fermentation time.Conclusion The method established in this study is simple,accurate,reproducible,and stable,and can be used for quality control of Massa Medicata Fermentata produced in Zhejiang Province.The optimum fermentation parameters of Massa Medicata Fermentata produced in Zhejiang Province were 30℃of temperature,80%of relative humidity,and 3 d of fermentation time.
