磷酸化聚酰胺-胺交联胶原蛋白支架体内成骨能力初探

摘要 目的:探究磷酸化聚酰胺-胺(P-PAMAM)树状大分子交联胶原蛋白支架在体内成骨效能。方法:通过傅里叶变换红外光谱(FTIR)对P-PAMAM进行表征,扫描电镜对胶原蛋白支架、聚酰胺-胺(PAMAM)复合胶原蛋白支架及P-PAMAM复合胶原蛋白支架进行形态表征。将3种复合支架植入SD大鼠颅骨骨缺损后,使用Micro-CT三维重建分析骨愈合过程中骨质、骨量的变化,采用HE染色及Masson染色对大鼠颅骨标本进行组织学分析。结果:FTIR显示PAMAM被成功磷酸化;扫描电镜显示,复合支架均具有良好的孔隙;体内成骨实验显示,与胶原蛋白支架和PAMAM复合胶原蛋白支架相比,P-PAMAM复合胶原蛋白支架具有更好的成骨诱导作用。结论:P-PAMAM树状大分子复合胶原蛋白支架在体内具有促进成骨作用。 Objective:This study investigated the osteogenic efficacy of phosphorylated polyamideamine(P-PAMAM)dendritic macromolecules cross-linked to collagen scaffolds in SD rats.Methods:Fourier transform infrared spectroscopy(FTIR)was used to characterize the phosphorylation of PAMAM and the morphological identification of three groups of collagen scaffolds,namely collagen scaffold group,PAMAM composite scaffold group and P-PAMAM composite scaffold group.Micro-CT three-dimensional reconstruction was used to analyze the changes of bone mass during bone healing after skull defect implantation in rats with composite scaffolds.HE staining and Masson staining were used to analyze the skull specimens of rats.Results:FTIR showed that PAMAM was successfully phosphorylated;scanning electron microscopy showed that the composite scaffold pore formation was good;and Micro-CT analysis of bone volume,ratio of new bone mass to total defect volume,bone mineral density and trabecular bone count,HE staining and Masson staining showed that the P-PAMAM composite collagen scaffold had better osteogenic induction effect than the collagen scaffold and PAMAM composite scaffold.Conclusions:P-PAMAM dendritic macromolecule composite collagen scaffold has a good effect on osteogenesis in vivo.
