金合欢素抑制人肺腺癌A549细胞生长及其作用机制的初步研究

Preliminary investigation on the inhibition of human lung adenocarcinoma A549 cell growth by acacetin and its mechanism of action

目的:探讨金合欢素对人肺腺癌A549细胞生长的影响及其可能的作用机制。方法:将A549细胞随机分组,采用CCK-8法计算不同浓度金合欢素作用下A549细胞的存活率。采用平板克隆实验反映金合欢素对增殖的影响。采用划痕实验检测金合欢素对A549细胞迁移的影响,采用Western Blot检测金合欢素作用下细胞中凋亡蛋白的表达。结果:CCK-8实验结果提示与对照组(0μmol/L)相比,A549细胞的存活率随着金合欢素浓度的升高而下降(P<0.05),差异有统计学意义;平板克隆实验结果发现与对照组(0μmol/L)相比,A549细胞的克隆数随着金合欢素浓度的升高而减少(P<0.05);划痕实验结果提示A549细胞的迁移率随着金合欢素浓度的升高而下降(P<0.05);Western Blot实验结果提示随着药物浓度的升高,与对照组(0μmol/L)相比,A549细胞中Bax蛋白的表达增多(P<0.05),Bcl-2蛋白和Survivin蛋白的表达减少(P<0.05)。结论:金合欢素可抑制A549细胞的增殖和迁移,诱导A549细胞凋亡。

Objective To investigate the effect of acacetin on the growth of human lung adenocarcinoma A549 cells and its possible mechanism.Methods A549 cells were randomly grouped,and the survival rate of A549 cells under the effect of different concentrations of acacetin was calculated by CCK-8 method.Plate cloning assay was used to reflect the effect of acacetin on proliferation.Results The results of CCK-8 assay suggested that the survival rate of A549 cells decreased with the increase of drug concentration(P<0.05)compared with the control group(0μmol/L),and the difference was statistically significant;The results of plate cloning assay revealed that the number of clone formation of A549 cells decreased with the increase of drug concentration(P<0.05)compared with the control group(0μmol/L);The results of the scratch assay suggested that the migration rate of A549 cells decreased with the increase of drug concentration(P<0.05).The results of the Western Blot assay suggested that with the increase of drug concentration,compared with the control group(0μmol/L),the expression of Bax protein increased in A549 cells(P<0.05),and the expression of Bcl-2 protein and Survivin protein decreased(P<0.05).Conclusions Acacetin can inhibit the proliferation and migration of A549 cells and induce apoptosis of A549 cells.
