超声辅助低共熔溶剂法提取太白贝母总黄酮及其抗氧化活性研究

Extraction of Total Flavonoids from Fritillaria taipaiensis by Ultrasonic-Assisted Deep Eutectic Solvent Method and Its Antioxidant Activity

摘要 采用超声辅助低共熔溶剂法提取太白贝母总黄酮,以太白贝母总黄酮提取率为考察指标,通过单因素实验和响应面法对提取工艺进行了优化,并通过DPPH自由基清除实验和ABTS自由基清除实验研究了其抗氧化活性。结果表明,在超声功率为69 W、料液比为1∶18(g∶mL)、超声温度为79℃、超声时间为27 min的最优工艺条件下,太白贝母总黄酮提取率可以达到17.7004%,与预测值相差极小;太白贝母总黄酮是一种强抗氧化剂,对DPPH自由基、ABTS自由基具有较强的清除能力,在浓度为16 mg·mL^(-1)时,清除率分别达到82.15%、90.42%,与VC无明显差异。超声辅助低共熔溶剂法操作简便、提取率高,适用于太白贝母总黄酮的提取。该研究为太白贝母药材资源的开发利用提供了参考。 We extracted the total flavonoids from Fritillaria taipaiensis by ultrasonic-assisted deep eutectic solvent method.Moreover,we optimized the extraction process through single-factor experiments and response surface methodologies by using the extraction rate of total flavonoids from Fritillaria taipaiensis as an evaluation index.Furthermore,we studied the antioxidant activity through DPPH free radicals and ABTS free radicals scavenging experiments.The results show that,under the optimal conditions as follows:the ultrasonic power of 69 W,the solid-liquid ratio of 1∶18(g∶mL),the ultrasonic temperature of 79℃,and the ultrasonic time of 27 min,the extraction rate of total flavonoids from Fritillaria taipaiensis reaches 17.7004%,which is very close to the predicted value.The total flavonoids from Fritillaria taipaiensis has strong scavenging ability against DPPH free radicals and ABTS free radicals.The total flavonoids from Fritillaria taipaiensis is a strong antioxidant.When the concentration of the total flavonoids from Fritillaria taipaiensis is 16 mg·mL^(-1),the scavenging rates will reach 82.15%and 90.42%,respectively,which is negligibly different from that of VC.The ultrasonic-assisted deep eutectic solvent method is suitable for the extraction of total flavonoids from Fritillaria taipaiensis with easy operation and high yield.This study provides a reference for the development and utilization of Fritillaria taipaiensis medicinal resources.
