摘要
目的研究肝硬化动物模型的造模特点,为肝硬化动物模型的规范化提供指导。方法通过检索中国知网、万方、维普、中国生物医学文献及PubMed数据库获取肝硬化动物模型文献,归纳实验动物种类、造模方法、给药方式、高频检测指标等构建数据库,并进行统计学分析。结果纳入符合标准的165篇文献,建立肝硬化动物模型以SD大鼠、Wistar大鼠和C57BL/6小鼠为主,大部分采用CCl 4+乙醇联用制备实验动物肝硬化模型,周期一般在5~8周,大鼠剂量以CCl 4-油混合液按每100 g体质量0.3 mL使用,乙醇溶液代替饮水的用法最为常见。给药方式中,CCl 4以皮下注射为主,+TAA以饮用为主。检测方式多以苏木精-伊红染色(HE)和全自动生化分析仪为主,主要检测肝脏组织病理学、肝组织或血清中的生化指标。结论肝硬化动物模型虽已成为当前肝病研究热点,但存在论述不全、说法笼统、阳性对照药的选择、以及缺乏中医证候的评价标准等问题。本文通过对造模情况进行统计分析,提出几点建议和设想,为肝硬化动物模型的规范化、客观化、标准化提供参考,便于开展肝硬化疾病的中医药研究。
Objective To study the modeling characteristics of animal models of liver cirrhosis and provide reference values for the standardization of animal models of liver cirrhosis.Method Literature on animal models of liver cirrhosis was obtained by searching China Knowledge Network,Wanfang,Vipshop,China Biomedical Literature and PubMed databases,and the database was constructed by summarizing the types of experimental animals,modeling methods,drug delivery methods,and high-frequency detection indexes,etc.,and statistical analysis was conducted.Results The 165 papers that met the criteria were included,and the animal models of cirrhosis were established mainly in SD rats,Wistar rats and C57BL/6 mice,and most of them used CCl 4(carbon tetrachloride)+ethanol combination to prepare experimental animal models of cirrhosis,with the cycle generally at 5~8 W,and the dose of rats was used at 0.3 mL per 100 g body weight with CCl 4-oil mixture.The ethanol solution instead of drinking water was the most common usage.Among the administration methods,CCl 4 was mainly injected subcutaneously and TAA was mainly drunk.The detection methods were mostly hematoxylin-eosin staining(HE)and fully automated biochemical analyzer,which mainly detected biochemical indexes in liver histopathology,liver tissue or serum.Conclusion Although animal models of liver cirrhosis have become a hot spot in current research on liver disease,there are problems such as incomplete discussion,generalized statements,selection of positive control drugs,and lack of evaluation criteria for TCM evidence.In this paper,through statistical analysis of the modeling situation,we proposed several suggestions and ideas to provide reference for the standardization,objectivity,and standardization of animal models of cirrhosis,and to facilitate the development of TCM research on cirrhotic diseases.
