摘要
目的:基于生物信息学研究方法分析TMPO基因在肺癌中的表达,评估其与肺癌诊断及预后评估方面的相关性。方法:以GTEx和TCGA数据库为发现数据库,获得健康人群以及肺癌患者的RNA-seq测序结果,绘制箱式图及配对点图展示TMPO基因在肺癌组织及正常组织中的差异化表达。绘制受试者曲线评估TMPO基因用于肺癌的诊断效能。根据TMPO基因表达水平的中位数将肺癌患者区分为TMPO低表达和高表达两组,研究患者的临床病理特征与TMPO基因表达水平的关系。使用Kaplan-MeierPlotter数据库绘制生存曲线,探究TMPO基因的高低表达与肺癌患者生存状况的关系。进一步通过单因素及多因素COX回归分析,筛选出TMPO表达水平为影响患者预后的独立危险因素。结果:肺癌患者的TMPO的表达水平为5.20(4.67~5.79),明显大于正常组织组中的4.29(4.04~4.63),差异具有统计学意义(P<0.05);同样的,在107对配对组织中,TMPO基因在肺癌组织中的表达水平5.50(4.78~6.16)也明显高于对应的癌旁正常组织TMPO的表达水平4.64(4.39~4.84),差异具有统计学意义(P<0.05);受试者曲线的结果表明TMPO用于诊断肺癌有着较高的敏感性[曲线下面积=0.842,95%置信区间为(0.822~0.862)];在针对TMPO基因表达和肺癌患者临床病理特征的关系研究中发现TMPO高表达的肺癌患者相比TMPO低表达的有着更晚的临床分期、T分期以及N分期,差异具有统计学意义(P<0.05);TMPO基因高表达的肺癌患者总体生存期以及无进展生存期也短于TMPO基因低表达的患者;此外,单因素及多因素COX回归分析,最终筛选出临床病理分期、T分期以及TMPO表达水平为影响患者预后的独立危险因素。结论:TMPO有潜力成为用于肺癌诊断和预后评估的新型生物学标志物。
Objective:To investigate the expression of the TMPO gene in lung cancer using bioinformatics research methods and evaluate its significance in the diagnosis and prognostic assessment of lung cancer.Methods:RNA-seq sequencing results were obtained from healthy populations and lung cancer patients using GTEx and TCGA databases as discovery databases.Box plots and paired dot plots were then utilized to illustrate the differential expression of TMPO genes in lung cancer tissues compared to normal tissues.Subject curves were plotted to evaluate the diagnostic efficacy of the TMPO gene for lung cancer.Patients with lung cancer were divided into two groups based on the median TMPO gene expression level,and the correlation between clinicopathological characteristics and TMPO gene expression level was examined.Survival curves were generated using the Kaplan-Meier Plotter database to investigate the correlation between high and low expression levels of the TMPO gene and the survival outcomes of patients with lung cancer.TMPO expression level was further screened as an independent risk factor affecting patients'prognosis by univariate and multivariate COX regression analyses.Results:The expression level of TMPO in lung cancer patients was 5.20(4.67~5.79),significantly higher than the level of 4.29(4.04~4.63)in normal tissue group,with a statistically significant difference(P<0.05).Similarly,in the 107 pairs of lung cancer tissues,TMPO gene expression was 5.50(4.78~6.16),significantly elevated compared to the expression level of TMPO in corresponding paracancerous normal tissues at 4.64(4.39~4.84),also statistically significant(P<0.05).The results of the receiver operating characteristic curves demonstrate a high sensitivity of TMPO in diagnosing lung cancer[area under the curve=0.842,95%confidence interval(0.822~0.862)].In the study examining the correlation between TMPO gene expression and clinicopathological characteristics in lung cancer patients,it was observed that individuals with high TMPO expression tended to have more advanced clinical stage,T stage,and N stage compared to those with low TMPO expression.These differences were found to be statistically significant(P<0.05).The overall survival and progressionfree survival of lung cancer patients with high TMPO gene expression were found to be shorter compared to those with low TMPO gene expression.In addition,univariate and multivariate COX regression analyses ultimately screened clinicopathological stage,T stage,and TMPO expression level as independent risk factors affecting patients'prognosis.Conclusion:TMPO has the potential to be a novel biomarker for the diagnosis and prognostic assessment of lung cancer.
