摘要
C-C基序趋化因子受体8(C-C motif chemokine receptor 8,CCR8)是一种调控免疫细胞趋化迁移的7次跨膜蛋白,在肿瘤微环境中高表达于Treg表面。由于CCR8^(+)Treg能强烈抑制肿瘤免疫,因此CCR8是一种扭转抑制型肿瘤微环境的理想靶点。目前有多种在研的靶向CCR8抗体药物,可在多种肿瘤模型中特异性清除肿瘤浸润CCR8^(+)Treg、扭转免疫抑制,展现出良好的抗肿瘤活性。此文对CCR8^(+)Treg促进肿瘤进展的基础研究和靶向药物应用进行综述,以期为该靶点肿瘤免疫治疗研究提供参考。
C-C motif chemokine receptor 8(CCR8)is a kind of seven-transmembrane protein that regulates the chemotaxis of immune cells,and is highly expressed on the surface of Tregs in the tumor microenvironment(TME).CCR8^(+)Tregs strongly inhibit tumor immunity,so CCR8 is an ideal target for reversing suppressing TME.At present,there are a variety of anti-CCR8 antibody drugs with prominent anti-tumor activity under development,which can specifically eliminate tumor-infiltrating CCR8^(+)Tregs and reverse immunosuppression in many tumor models.This review summarizes the basic researches on CCR8^(+)Tregs promoting tumor progression and the application of targeted drugs,in order to provide a reference for tumor immunotherapy study.
