摘要
目的探究Salusin-β与NF-κB通路在2型糖尿病性干眼病炎症反应中的作用。方法将30只健康雄性Wistar大鼠随机分为空白对照组、阴性对照组、Salusin-β低表达组,每组10只。HE染色观察大鼠眼结膜的病理结构变化;酶联免疫吸附法测定泪液炎症因子的水平;蛋白免疫印迹实验分析大鼠结膜组织Salusin-β和NF-κB p65蛋白的表达。结果与阴性对照组比较,低表达组的大鼠眼表病变情况显著减轻、泪液分泌增加、角膜增厚、炎症细胞浸润和基质水肿的情况均得到改善、泪液中炎症因子显著降低、眼结膜Salusin-β和NF-κB p-p65蛋白表达水平显著降低。结论Salusin-β可能通过调控NF-κB通路促进2型糖尿病性干眼病的炎症反应。
Objective To investigate the role of Salusin-βand NF-κB pathway in the inflammation of type 2 diabetic dry eye.Methods Thirty healthy male Wistar rats were randomly divided into blank control group,negative control group and Salusin-βlow expression group,10 in each group.The pathological changes of rat conjunctiva were observed by HE staining.The levels of inflammatory factors in tears were measured by enzyme-linked immunosorbent assay.The expressions of Salusin-βand NF-κB p65 protein in rat conjunctiva was analyzed by Western blot.Results Compared with the negative control group,the ocular surface lesions of the rats in the Salusin-βlow expression group were significantly alleviated,the tear secretion was increased,the corneal thickening,inflammatory cell infiltration and matrix edema were improved,the inflammatory factors in tears were significantly reduced,and the levels of Salusin-βand NF-κB p-p65 in conjunctiva were significantly decreased.Conclusion Salusin-βmight promote inflammation in type 2 diabetic dry eye disease by regulating the NF-κB pathway.
