摘要
‘聊红’椿是本课题组从一种臭椿(Ailanthus altissima)芽变株经过多年培育而成的红果臭椿新品种(证书号为20190262),具有较高的观赏价值。建立‘聊红’椿组培快繁体系不仅有利于该品种迅速推广,同时也对研究果荚成色机理具有帮助。选取‘聊红’椿侧枝腋芽及顶芽(单芽)为外植体,研究了消毒方式、不同激素配比对‘聊红’椿愈伤形成、继代培养、不定芽形成和生根的影响,结果表明,以腋芽及顶芽(单芽)为外植体的消毒方式为依次采用75%乙醇浸泡30 s、15%次氯酸钠溶液15 min、0.1%氯化汞溶液6 min,组织污染率为20.00%,愈伤组织诱导成功率达40.00%;脱分化诱导和继代培养的MS培养基中激素配比为0.1 mg/L6-BA、0.1 mg/L NAA、2,4-D等量混合液,愈伤组织诱导率达到40.00%,愈伤组织颜色为黄绿色,长势良好;最适发芽培养基MS中激素配比为3.0 mg/L 6-BA、0.05 mg/L NAA、2,4-D等量混合液,不定芽诱导率达到82.33%;最适生根培养基MS的激素含量为NAA 2.0 mg/L,不定根诱导率为58.33%。‘聊红’椿的组织培养研究,可以满足‘聊红’椿快繁需求,具有生产容易、成本低、易操作、繁殖系数大等优点,本研究结果能够为‘聊红’椿的工厂化育苗提供一定技术支撑。
The new variety of'Liaohong'Ailanthus altissima(Certificate No.20190262)was bred from a bud strain of Ailanthus altissima by us.The establishment of the tissue culture and rapid clonal propagation system of'Liaohong'Ailanthus altissima is not only beneficial to popularize the variety rapidly,but also helpfull the study of the formation mechanism of pod color.The effects of disinfection methods,different hormone ratio on the culture medium MS of callus formation,subculture,adventitious bud formation and rooting of'Liaohong'Ailanthus altissima were studied,through selecting axillary buds and top buds as explants.The results showed that the disinfection methods of axillary buds and top buds were soaked in 75%alcohol for 30 s,15%sodium hypochlorite solution for 15 minutes,and then 0.1%mercuric chloride solution for 6 minutes,the tissue pollution rate was20.00%,and the success rate of callus induction was 40.00%;the ratio of hormone in MS medium for dedifferentiation induction and subculture was 0.1 mg/L 6-BA,0.1 mg/L NAA+2,4-D,and the induction rate of callus reached 40.00%,and the callus grow better;the optimal ratio of hormone in MS for adventitious bud formation was 3.0 mg/L 6-BA,0.05 mg/L NAA+2,4-D,and the induction rate of adventitious buds reached 82.33%;the optimal medium MS for rooting was NAA 2.0 mg/L and the induction rate of adventitious root was 58.33%.The tissue culture of'Liaohong'Ailanthus altissima can meet the needs of rapid propagation of'Liaohong'Ailanthus altissima.It has the advantages of easy production,low cost,easy operation and large propagation coefficient.The results of this study can provide some technical support for industrial nursery of'Liaohong'Ailanthus altissim.
