摘要
从诺丽种子中分离纯化得到一个新的倍半木脂素moricitan A(1),一个新天然产物4-甲氧基异美洲素A(2),一个新的蒽醌2,4-二羟基-3,8-二甲氧基-7-(甲氧基甲基)-9,10-蒽二酮(7)和7个已知的化合物3~6和8~10,其结构通过多种现代波谱技术进行确定,化合物1的绝对构型通过圆二色谱(ECD)计算确定.评价了所有化合物对脂多糖(LPS)诱导小鼠巨噬细胞RAW264.7产生一氧化氮(NO)的抑制作用,化合物1和9'-ethyl americanol A(3)表现出较好的抗炎活性,其IC_(50)分别为(2.52±0.22)和(5.46±0.89)μmol/L,强于阳性对照地塞米松[IC_(50)为(65.39±1.02)μmol/L].
A new sesquineolignan moricitan A(1),a new natural product 4-methoxy isoamericanin A(2),a new anthraquinone 1,3-dihydroxy-2,5-dimethoxy-6-(methoxymethyl)anthracene-9,10-dione(7),and seven known compounds 3~6 and 8~10 were obtained from the seeds of Noni.The structures were elucidated using comprehensive spectroscopic methods,and the absolute configuration of 1 was established based on quantum-chemical electronic circular dichroism calculations.The inhibitory activity of all the compounds on NO production in lipopolysaccharide-induced(LPS)mouse leukemia cells of monocyte macrophage(RAW 264.7)cells was evaluated.Compound 1 and 9'-ethyl americanol A(3)exhibited stronger anti-inflam-matory activity in vitro with IC_(50) values of(2.52±0.22)and(5.46±0.89)μmol/L to that of dexamethasone[IC_(50)=(65.39±1.02)μmol/L],respectively.
