摘要
树参在药用﹑食用和园林绿化等方面都有良好的发展前景,常规育苗方法已无法满足树参苗木的市场需求。为此,研究了树参组织培养技术,以期为以后的工厂化育苗提供理论基础。选择生长健壮的一年生半木质化、无病虫害的枝条为外植体,用单因素试验设计和正交试验设计的方法,研究了不同的培养基类型和激素浓度对树参组织培养再生过程的影响。结果表明:MS培养基比WPM培养基更适合树参不定芽的诱导,最佳培养基为MS+ZT1.5mg/L+6-BA1.5mg/L+NAA0.4mg/L;改良MS+6-BA1.0mg/L+KT0.6mg/L+NAA0.5mg/L的增殖率和增殖系数均最高;生根培养的基本培养基选用1/2MS,最佳培养基为1/2MS+IBA1.0mg/L+ABT 10.6mg/L+NAA0.1mg/L;泥炭土∶黄心土∶珍珠岩(2∶1∶1)适宜作为树参组培苗移栽基质,组培苗移栽成活率达89.8%。从而得出以下结论:高浓度的无机盐离子更能促进不定芽的萌发,但同时也增加了玻璃化的可能性;细胞分裂素ZT和6-BA在不定芽的诱导和增殖中起着重要的作用;高浓度的6-BA对不定芽的增殖会有抑制作用;生长素NAA对不定根的形成影响较小。
Dendropanax dentiger has a good development prospect in the fields of medicine,food and landscaping.Conventional seedling raising method has been unable to meet the market demand of Dendropanax dentiger seedlings.In this paper,the tissue culture technology of Dendropanax dentiger was studied in order to provide a theoretical basis for the factory seedling cultivation in the future.Healthy annual semi-lignified branches free of pests and diseases were selected as the explants.The effects of different medium types and hormone concentrations on the regeneration process of Dendropanax dentiger tissue culture were studied by means of single factor and orthogonal design.The test results show that:MS medium was more suitable for the induction of adventitious buds than WPM medium.The best medium is MS+ZT1.5mg/L+6-BA1.5mg/L+NAA0.4mg/L;The multiplication rate and multiplication coefficient of improve MS+6-BA1.0mg/L+KT0.6mg/L+NAA0.5mg/L were the highest.The basic medium for rooting culture was 1/2MS.The best medium is 1/2MS+IBA1.0mg/L+ABT10.6mg/L+NAA 0.1 mg/L;Peat soil∶yellow soil∶perlite(2∶1∶1)was suitable as the transplanting medium,and the survival rate of transplanting was 89.8%.The following conclusions can be drawn:High concentration of inorganic salt ions can promote the germination of adventitious buds,but also increase the possibility of glass transition;cytokinins ZT and 6-BA play important roles in the induction and proliferation of adventitious buds;high concentration of 6-BA can inhibit the proliferation of adventitious buds;auxin NAA had little effect on adventitious root formation.
