茶树CsTRPS基因的鉴定及豌豆-茶树间作条件下表达分析

摘要 TRPs(tryptophan synthase beta chain 1)基因调控合成的色氨酸,作为吲哚乙酸合成的前体,广泛存在于各种植物中,并通过调控生长素的合成影响植物的生长。但关于茶树中TRPs基因研究尚未报道。因此,以茶树为试验材料,克隆了茶树CsTRPS基因,对CsTRPS基因进行了生物信息学分析,并通过转录组数据探究了豌豆-茶树间作条件下基因的表达情况。结果表明:CsTRPS基因全长1425bp,编码474个氨基酸,CsTRPS蛋白分子量为51.552865KDa,理论等电点(PI)为6.52,甘氨酸(Gly)残基含量最高,占比10.1%,为亲水性蛋白,无信号肽及跨膜结构。CsTRPS蛋白二级结构中α-螺旋占比38.19%,β-转角占比7.59%,延伸链占比17.09%,无规则卷曲占比37.13%。亚细胞定位预测结果表明,CsTRPS蛋白主要集中在线粒体中表达。系统进化树显示,茶树CsTRPS蛋白与红马银花RvTRPS蛋白,猕猴桃AcTRPS蛋白,君迁子DlTRPS蛋白进化关系最近。转录组数据显示,茶树CsTRPS基因在豌豆-茶树间作条件下下调表达。 TRPs(tryptophan synthase beta chain 1)gene,as a precursor to the synthesis of indole acetic acid,regulated the synthesis of tryptophan,which is widely existed in various plants,and affected the growth of plants by regulating the synthesis of auxin.However,studies on TRPs genes in tea plants have not been reported.Therefore,this study took tea tree as experimental material,cloned CsTRPS gene of Camellia sinensis,and analyzed CsTRPS gene bioinformatically.The transcriptome data were used to investigate the gene expression in the pea-Camellia sinensis intercropping.The results showed that the total length of CsTRPS gene was 1425 bp,encoding 474 amino acids.The molecular weight of CsTRPS protein was 51.552865 KDa;the theoretical isoelectric point(PI)was 6.52;the content of glycine(Gly)residue was the highest,accounting for 10.1%,and it was a hydrophilic protein without signal peptide and transmembrane structure.In the secondary structure of CsTRPS protein,α-helix accounted for 38.19%;β-angle accounted for 7.59%;extended chain accounted for 17.09%,and irregular curling accounted for 37.13%.Subcellular localization prediction showed that CsTRPS protein was mainly expressed in mitochondria.Phylogenetic tree showed that CsTRPS protein of Camellia sinensis had the closest evolutionary relationship with RvTRPS protein of Rhododendron vialii Delavay et Franch.,AcTRPS protein of kiwifruit,and DlTRPS protein of Diospyros lotus L..Transcriptome data showed that the expression of CsTRPS gene in Camellia sinensis plant was down-regulated under pea-Camellia sinensis intercropping conditions.
