摘要
采用H_(2)O_(2)脱色和AB-8大孔吸附树脂柱层析法分别对溶剂提取的粗皂荚皂素(GSS)进行分离纯化,对比研究了其纯度、结构组成、表面活性、泡沫及乳化性能。研究结果表明:H_(2)O_(2)脱色纯化皂素(H_(2)O_(2)-GSS)的皂苷元结构发生了破坏,皂苷质量分数为48.02%,但其稳泡性能优异,泡沫半衰期(t_(1/2))为600 min(10 h);柱层析法中60%乙醇洗脱皂素(60%-GSS)纯度最高,皂苷质量分数达到92.12%,泡沫性能最优,泡沫高度达11 cm,乳液在24 h内最稳定,在2.2 cm处发生分层;75%乙醇洗脱皂素(75%-GSS)皂苷质量分数为86.40%,其表面张力值最低,为41.50 mN/m,临界胶束浓度(C_(CMC))值为3 g/L,表面活性最好。H_(2)O_(2)-GSS具有较强的稳泡能力,柱层析纯化法分离得到的皂荚皂素纯度更高,表面活性、泡沫性能和乳液稳定性更好,纯化重组皂素(M-GSS)表面活性进一步提高,为高性能表面活性剂的开发提供了思路。
Commercially available G.sinensis saponin(GSS)had deep color,heavy odor,and low saponin content,which limited its application.In this paper,the solvent extraction of GSS was separated and purified by H_(2)O_(2)decolorization and AB-8 macroporous adsorption resin column chromatography.The purity,structure composition,surface activity,foam and emulsification properties of saponin were compared and studied.The results showed that the structure of saponin purified by H_(2)O_(2)decolorization(H_(2)O_(2)-GSS)was damaged,the saponin content was 48.02%,but the foam stability was excellent and the half-life(t_(1/2))was 10 h.Results of column chromatography showed that the highest purity of saponin was achieved through elution using 60%ethanol,resulting in a 92.12%saponin mass fraction.Additionally,the foam performance was the best,reaching a height of up to 11 cm,and the emulsion was the most stable within 24 h.After 24 hours,stratification occurred at a height of 2.2 cm.The mass fraction of saponin eluted by 75%ethanol was 86.40%,the surface tension value was the lowest(41.50 mN/m),and the critical micelle concentration(C_(CMC))was 3 g/L(having the best surface activity).The H_(2)O_(2)-GSS showed better foam stabilization ability.The GSS obtained by column chromatography had higher purity,better surface activity,foam performance and emulsion stability.The surface activity of purified recombinant saponin(M-GSS)was further improved,which provided ideas for the development of high-performance surfactants.
