Expression and clinical significance of GLIS3 in hepatocellular carcinoma

目的 探讨锌指蛋白GLIS3在肝细胞肝癌(HCC)中的表达及其临床意义。方法 收集TCGA、GTEx和GEO等数据库中GLIS3在肝细胞肝癌中的表达值,对2101例HCC组织和1448例正常肝组织进行综合分析,利用实时定量逆转录聚合酶链反应技术检测肝细胞癌细胞系(MHCC-97H、SK-HEP-1)和正常肝细胞细胞系(LX2)中GLIS3 mRNA的表达,使用免疫组化技术检测96例HCC组织及其相邻的96例非癌组织中GLIS3蛋白的表达水平。结果 数据库分析结果显示,GLIS3在癌组织中的表达明显低于非癌组织(P<0.001)。GLIS3在MHCC-97H、SK-HEP-1肝细胞癌细胞系中的表达明显低于正常肝细胞细胞系(t=2.933,P<0.05;t=11.892,P<0.05)。GLIS3蛋白表达在不同HCC患者肿瘤数目方面差异具有统计学意义(P<0.05)。GLIS3与HCC患者预后相关(P<0.05)。结论 GLIS3在肝细胞肝癌组织中表达较低,可能成为肝细胞癌治疗潜在的靶点、诊断和预后判断的标记物。

Objective To evaluate the expression and clinical relevance of zinc finger protein GLIS3 in hepatocellular carcinoma(HCC).Methods Expression data for GLIS3 were extracted from TCGA,GTEx,and GEO databases for analysis involving 2101 HCC and 1448 normal liver samples.GLIS3 mRNA expression was quantified in HCC cell lines(MHCC-97H,SK-HEP-1) and a normal liver cell line(LX2) using real-time quantitative PCR.GLIS3 protein expression levels were assessed via immunohistochemistry in 96 paired samples of HCC and adjacent non-tumor tissue.Results The results of database analysis showed that GLIS3 expression was significantly lower than that in HCC tissues compared to normal liver tissue(P<0.001).Lower GLIS3 expression was confirmed in MHCC-97H and SK-HEP-1 HCC cell lines relative to the normal liver cell line(t=2.933,P<0.05;t=11.892,P<0.05).There is statistically significant difference between GLIS3 protein expression and tumor number in HCC patients.GLIS3 was associated with prognosis in HCC patients.Conclusion Reduced GLIS3 expression in HCC suggests its potential as a therapeutic target and prognostic biomarker.
