摘要
目的 合成明胶/羟丙基-β-环糊精/甘油-水复合凝胶,探讨羟丙基-β-环糊精对该凝胶体系的影响及凝胶载药应用。方法 采用溶胶-凝胶法制备凝胶,并使用多种物理方法如扫描电子显微镜(SEM)、差示扫描量热法(DSC)、傅里叶红外光谱(IR)、X射线衍射(XRD)、流变学等方法对复合凝胶进行表征;以牛津杯法验证该凝胶递送左氧氟沙星的抗菌效果。结果 羟丙基-β-环糊精以氢键结合明胶,促进明胶分子螺旋微结构聚集,增强了复合凝胶的稳定性,流变测试可知该复合凝胶应力屈服值增大了1000Pa。复合凝胶装载抗菌药物左氧氟沙星后作用于金黄色葡萄球菌和大肠埃希氏菌,抑菌效能可分别达到对照标准的90.27%和80.02%。结论 新型复合凝胶合成方法简便、易行,体系中的羟丙基-β-环糊精和甘油都可改善复合凝胶的微结构、性质和性能,该复合凝胶的左氧氟沙星制剂表现出极好的抑菌效果。
Objective A gelatin(GA) gel modified by hydroxypropyl β-cyclodextrin(HPCD) will be synthesized,and its micro-structure,physical properties,antibacterial effect of loaded-drug levofloxacin(LVFX) will also be investigated.Methods The composite gel was synthesized by sol-gel method,and its structure and properties were studied by OM,SEM,FTIR,XRD and rheology.The antibacterial effect of the composite gel loaded with levofloxacin was tested by Oxford cup method.Results The GA spiral molecules could be collected by H-bond force of HPCD,which enhanced the stability of the composite gel network and increased the stress yield value of 1000 pa.The antibacterial efficacy of the loaded-drug gel with LVFX could reach about 90.27% and 80.02% to the positive antibacterial control on Staphylococcus aureus and Escherichia coli,respectively.Conclusion The composite gel can be acquired in a direct and easy method.The microstructure and properties of GA/HPCD gel were optimized by H-bonding interactions between GA and HPCD or glycerol molecules.The composite gel deserves to be widely applied as drug delivery system.
