摘要 目的:探讨ALKBH5在结直肠癌(colorectal cancer, CRC)及其癌旁组织中的差异表达,评估其作为CRC诊断肿瘤标志物的潜在应用。方法:通过GEPIA2数据库和UALCAN网站检索ALKBH5在CRC和癌旁组织中的表达水平,结合PCR和琼脂糖凝胶电泳验证ALKBH5 cDNA引物的特异性,并通过RT-qPCR分析ALKBH5的熔解温度曲线图。结果:ALKBH5在不同癌症中的表达水平和作用功能存在差异,在CRC中的相对表达量较低,而在癌旁组织中则较高,但与患者预后无显著相关性。PCR产物片段大小与ALKBH5 cDNA的121 bp一致,电泳条带清晰且单一,表明引物特异性良好。熔解温度曲线图显示RT-qPCR产物特异性良好,未产生引物二聚体。AUC值为0.945 8,表明ALKBH5具有较高的辅助诊断效能。结论:ALKBH5有望作为CRC诊断的肿瘤标志物,但ALKBH5 mRNA表达量难以单独作为CRC患者治疗靶点,需要综合考虑其表达产物的多样性。 Objective:To investigate the differential expression of ALKBH5 in colorectal cancer(CRC)and para-cancerous tissues to evaluate its potential as a diagnostic biomarker for CRC.Methods:The expression levels of ALKBH5 in CRC and para-cancerous tissues were retrieved through the GEPIA2 database and UALCAN website.The specificity of ALKBH5 cDNA primers was validated by PCR and agarose gel electrophoresis,and the melting temperature curves of ALKBH5 were analyzed by RT-qPCR.Results:The expression levels and functional roles of ALKBH5 vary among different cancers.The relative expression level is lower in CRC and higher in para-cancerous tissues,with no significant correlation to patient prognosis.The PCR product size was consistent with the 121 bp fragment of the ALKBH5 cDNA,and the electrophoresis bands were clear and singular,indicating excellent primer specificity.The melting temperature curve analysis showed good specificity of the RT-qPCR products without the presence of primer dimers.An AUC value of 0.9458 indicates a high diagnostic efficacy of ALKBH5.Conclusion:ALKBH5 shows promise as a diagnostic biomarker for CRC.However,the expression level of ALKBH5 mRNA alone is insufficient to serve as a therapeutic target for CRC patients.It is necessary to consider the diversity of its expression products comprehensively.
