钛表面ZIF-L涂层的构建及细胞毒性检测

Construction and cytotoxicity test of ZIF⁃L coating on titanium

摘要 目的在钛(titanium,Ti)表面构建新型锌基金属有机骨架配合物(Ti@ZIF-L)涂层并进行生物学评价。方法通过原位生长法在碱热处理后Ti试件表面构建锌基金属有机骨架材料(Ti@ZIF-L)。利用扫描电镜观察材料表面形貌变化;接触角测量仪检测Ti和Ti@ZIF-L表面接触角;扭矩实验检测Ti@ZIF-L涂层的结合强度;利用细胞毒性实验评价Ti@ZIF-L的细胞相容性。结果扫描电镜观察到Ti@ZIF-L表面形成多级、花状的纳米结构;接触角检测结果显示:Ti@ZIF-L接触角为38.11°,与未处理的Ti相比,接触角降低26.22°(P<0.05);扭矩实验结果显示,在扭矩为9.48 N·m时,扫描电镜仍可见Ti表面生长的三维结构的ZIF-L,Ti与ZIF-L涂层间结合良好;细胞毒性实验显示,Ti与Ti@ZIF-L在不同浓度浸提液之间差异无统计学意义(P>0.05)。结论Ti@ZIF-L涂层材料具有良好的稳定性和细胞相容性,有望应用于临床中对种植材料的表面改性。 Objective To construct a novel zinc⁃based coating with a metal⁃organic framework complex(Ti@ZIF⁃L)on a titanium(Ti)surface and conduct the biological evaluation.Meth⁃ods Zinc⁃based metal⁃organic framework material was constructed on the surface of the Ti specimen after alkali heat treatment by in⁃situ growth method(Ti@ZIF⁃L).The surface mor⁃phology of the coating was observed by scanning electron microscopy(SEM),the surface con⁃tact angles of Ti and Ti@ZIF⁃L were measured by contact angle measuring instrument,and the bonding strength of Ti@ZIF⁃L was measured by torque test;The cytocompatibility of Ti@ZIF⁃L was evaluated by cytotoxicity assay.Results Multistage flower⁃like nanostructures were found on the surface of Ti@ZIF⁃L by SEM;The contact angle test results showed that the con⁃tact angle of Ti@ZIF⁃L was 38.11°,which was 26.22°lower than that of untreated Ti(P<0.05);The results of the torque experiment showed that the three⁃dimensional structure of ZIF⁃L growing on the surface of Ti could still be seen by SEM when the torque was 9.48 N·m,in⁃dicating that the combination between Ti and ZIF⁃L coating was good;The cytotoxicity test showed that there was no statistical difference between Ti and Ti@ZIF⁃L at different concentrations(P>0.05).Conclusion Ti@ZIF⁃L coating material has good stability and cell compati⁃bility and is expected to be used in clinical surface modification of implant materials.
