冠心病患者唾液菌群结构和组成研究

摘要 目的 探讨冠心病患者唾液菌群结构和组成。方法 回顾性收集2022年5至6月温岭市第一人民医院收治的10例冠心病患者以及19名同期本院健康体检者唾液样本,分别设为冠心病组、对照组。提取唾液样本总DNA,采用16S r DNA高通量测序技术检测唾液菌群结构和组成,比较两组研究对象唾液菌群相对丰度以及数据α多样性指数(包括Chao指数、ACE指数、Shannon指数、Simpson指数)。结果 门分类分析显示,两组研究对象唾液菌群相对丰度比较差异均无统计学意义(均P>0.05),但个体间差异大。属分类分析显示,冠心病组唾液韦荣球菌属相对丰度明显高于对照组(P<0.001),且个体间差异大;而嗜血杆菌属、密螺旋体属、沃林氏菌属相对丰度均明显低于对照组(均P<0.05)。种分类分析显示,冠心病组唾液罗伊氏乳杆菌、殊异韦荣球菌相对丰度均明显高于对照组(均P<0.01),但前者丰度较低,且个体间差异大。两组研究对象数据Chao指数、ACE指数、Shannon指数、Simpson指数比较,差异均无统计学意义(均P>0.05)。结论 冠心病患者唾液殊异韦荣球菌明显升高,是特征性表达的口腔菌群。 Objective To explore the structure and composition of saliva microbiota in patients with coronary heart disease.Methods The saliva samples of 10 patients with coronary heart disease and 19 healthy subjects in Wenling first people's Hospital from May to June 2022 were collected and designated as the coronary heart disease(CHD)group and the control group.Total DNA was extracted from saliva samples,and 16S rDNA high-throughput sequencing technique was used to detect the structure and composition of salivary microbiota.The relative abundance of salivary microbiota and dataαdiversity indices(including Chao index,ACE index,Shannon index,Simpson index)were compared between the two groups.Results The results of the cluster analysis indicated that there was no significant difference in the relative abundance of salivary microbiota between the two groups(all P>0.05),but there were significant differences among individuals.Genus taxonomic analysis showed that the relative abundance of Streptococcus in the saliva of the CHD group was significantly higher than that of the control group(P<0.001),and there were great differences among individuals.The relative abundance of Haemophilus,treponema and Veillonella were significantly lower in the CHD group compared to the control group(all P<0.05).The Species cluster analysis showed that the relative abundance of lactobacillus reuteri and veillonococcus aberrans in the saliva of the CHD group was significantly higher than that of the control group(all P<0.01),but the abundance of the former was lower and the differences among individuals were large.There was no significant difference in Chao index,ACE index,Shannon index and Simpson index between the two groups(all P>0.05).Conclusion Salivary levels of Streptococcus sanguinis were significantly elevated in CHD patients,representing a characteristic expression of oral microbiota.
