艾灸神阙穴对原发性痛经模型大鼠的作用及其机理

The Effect and Mechanism of Moxibustion Treatment on a Primary Dysmenorrhea Rat Model at the"Shenque"(CV 8)

目的:探究艾灸神阙穴对原发性痛经模型大鼠的作用及其机制。方法:通过阴道涂片实验确定大鼠动情周期后使用苯甲酸雌二醇联合缩宫素制备原发性痛经大鼠模型。通过观测原发性痛经大鼠体质量、脏器指数、扭体潜伏期和扭体分数变化,检测各组大鼠血清性激素、前列腺素和疼痛因子等各项指标水平,比较艾灸治疗原发性痛经大鼠药效差异,其艾灸种类包括有烟艾灸与无烟艾灸。结果:在大鼠一般情况和行为学测试观察中,模型组比空白组扭体反应显著增加(P<0.01),表明造模成功;而与模型组比较,两个艾灸组大鼠体质量减轻相对缓慢,扭体潜伏期显著增加,扭体反应显著减少(P<0.05),其中有烟艾灸组和无烟艾灸组、阳性药物组大鼠扭体抑制率分别为54.89%、37.21%、68.82%。与模型组比较,两个艾灸组大鼠血清中雌二醇、缩宫素、前列腺素F_(2α)、白介素-6浓度均显著降低(P<0.05),前列腺素E_(2)和β-内啡肽水平显著升高(P<0.05)。结论:艾灸大鼠神阙穴能够有效缓解原发性痛经,调节机体性激素、前列腺素和疼痛相关因子可能是其潜在的作用机制。

Objective:To investigate the therapeutic effects and mechanisms of moxibustion on the"Shenque"(CV 8)in primary dysmenorrhea rat model.Methods:A primary dysmenorrhea rat model was established using estradiol benzoate and oxytocin after determining the estrous cycle through vaginal smear tests.Changes in rat body weight,organ index,writhing latency and writhing scores were observed.Various indicators,including serum hormones,prostaglandins and pain-related factors,were measured to compare the efficacy of moxibustion in treating primary dysmenorrhea in rats.Moxibustion methods included smokeless moxibustion and moxibustion with smoke.Results:In general conditions and behavioral tests of rats,the model group showed a significant increase in writhing response compared to the blank group(P<0.01),indicating a successful modeling.Compared to the model group,both moxibustion groups exhibited slower reductions in body weight,a significant increase in writhing latency and a significant decrease in writhing response(P<0.05).The writhing inhibition rates in the smokeless moxibustion,moxibustion with smoke and positive drug groups were 54.89%,37.21%and 68.82%,respectively.Compared to the model group,both moxibustion groups showed significant reductions in serum concentrations of estradiol,oxytocin,prostaglandin F_(2α),and interleukin-6(P<0.05),while prostaglandin E_(2)andβ-endorphin levels increased significantly(P<0.05).Conclusion:Moxibustion on the"Shenque"(CV 8)can effectively alleviate primary dysmenorrhea in rats,and the modulation of endocrine hormones,prostaglandins,and pain-related factors may be its potential mechanisms of action.
