樱桃谷肉鸭肝脏发育早期IGF和IGF-ⅠR基因表达规律研究

摘要 樱桃谷肉鸭是世界著名的瘦肉型鸭,其肝脏发育对机体健康及生长至关重要。该研究旨在探讨樱桃谷肉鸭在胚胎后期和出壳早期肝脏发育及关键基因表达规律。试验选用120枚受精鸭蛋,在第16、21、28胚龄,和第7、14、21日龄记录胚体重、体质量重和肝脏质量重的变化。采用荧光定量PCR技术检测肝脏发育相关IGF1、IGF2、IGF-ⅠR mRNA的表达情况。结果表明,IGF1、IGF2、IGF-ⅠR在16胚龄鸭肝脏中已有少量表达。随胚龄/日龄增加IGF1 mRNA表达量呈现持续增长的态势,IGF2和IGF-ⅠR mRNA表达量呈现先增加后下降的趋势。IGF1、IGF2、IGF-ⅠR mRNA表达量相互间存在显著正相关,三个基因在鸭早期肝脏生长发育过程中存在协同表达现象,肝脏IGF1、IGF2、IGF-ⅠR mRNA的表达在鸭早期发育过程中发挥着重要作用。 Cherry Valley broiler duck is a world-renowned lean duck,and its liver development is crucial for the health and growth of the organism.The aim of this study was to investigate the liver development and key gene expression patterns of Cherry Valley ducks in the late embryonic and early emergence stages using the Cherry Valley duck as an experimental model.One hundred and twenty fertilized duck eggs were used in the experiment,and the changes in body weight and liver weight were recorded at the 16th,21st,and 28th embryonic ages,and at the 7th,14th,and 21st day of age.The expression of IGF1,IGF2,and IGF-ⅠR mRNAs related to liver development were detected by fluorescence quantitative PCR.The results showed that IGF1,IGF2 and IGF-ⅠR had been expressed in small amounts in the livers of ducks at 16 embryonic days of age.The expression of IGF1 showed a continuous increase with the increase of embryonic age/age,IGF2 and IGF-ⅠR showed a trend of increasing and then decreasing.There was a significant positive correlation among IGF1,IGF2 and IGF-ⅠR,and there was a synergistic expression phenomenon of the three genes in the early development of ducks'liver.The expression of hepatic IGF1,IGF2 and IGF-ⅠR mRNA played an important role in the early development of ducks.
