摘要
为探究阿克苏冰糖心苹果对小鼠肠道菌群的影响,给4组5周龄雄性昆明小鼠每天分别灌胃阿克苏冰糖心苹果糖心部分、阿克苏冰糖心苹果果肉(非糖心部分)、山梨醇以及生理盐水。21 d后在无菌条件下解剖取出小鼠结肠段,通过对结肠段的16S rRNA进行选择性扩增并用Illumina Novaseq 6000对扩增的基因序列进行高通量测序,对小鼠肠道菌群的多样性、丰富度、菌群物质组成结构以及功能基因代谢预测分析。结果表明,灌胃冰糖心苹果并未对正常小鼠肠道菌群丰富度与多样性产生显著变化。灌胃冰糖心苹果果肉(非糖心部分)使小鼠肠道变形菌门显著增加,也上调了控制新陈代谢的基因;灌胃冰糖心苹果糖心部分使小鼠肠道拟杆菌纲显著减少,芽孢杆菌纲显著增加;灌胃山梨醇可在一定程度上抑制变形菌门菌群的增长。该结果为进一步探索阿克苏冰糖心苹果更为深层次的内在价值提供了理论依据。
In order to investigate the impact of Aksu-grown candy apple on the intestinal microbiota of mice,four groups of 5-week-old male Kunming mice were orally administered daily with candy core of Aksu-grown candy apple,Aksu-grown candy apple flesh(non-candy core),sorbitol,and physiological saline,respectively.After 21 days,the mice's colon segments were dissected under sterile conditions,selective amplification of the colon segments'16S rRNA was performed,and the amplified gene sequences were subjected to high-throughput sequencing using Illumina Novaseq 6000 to analysis the diversity,richness,microbial composition,and functional gene metabolism prediction of the mice's intestinal microbiota was conducted.The results indicated that oral administration of Aksu-grown candy apple did not significantly alter the richness and diversity of the normal mouse intestinal microbiota.However,administration of Aksu-grown candy apple flesh(non-candy core)significantly increased the abundance of Proteobacteria phylum,accompanied by an upregulation of genes controlling metabolism.Administration of Aksu-grown candy apple candy core led to a significant reduction in Bacteroidales class and a notable increase in Bacilli class.Oral administration of sorbitol may to some extent inhibit the growth of Proteobacteria.These findings provide a theoretical basis for further exploring the intrinsic value of Aksu-grown candy apple at a deeper level.
