鲜葛中葛根素含量分布及淀粉和葛根素同步提取工艺

Distribution of Puerarin Content in Fresh Pueraria thomsonii Benth and Simultaneous Extraction Technology of Starch and Puerarin

摘要 葛根素是葛根的主要活性成分,但在葛根淀粉生产过程中,大量的葛根素随着葛粉加工废水流失,造成了资源浪费。在葛根淀粉提取时同步回收废水中的葛根素是实现葛根资源高值化利用的重要途径。文章采用高效液相色谱法分析葛根不同部位,以及葛粉提取过程废水中葛根素的含量分布规律,采用单因素试验及正交试验设计进行葛根淀粉和葛根素同步提取工艺优化。结果表明:超过80%的葛根素集中在葛皮中,第一次淀粉提取废水中葛根素含量占总废水的70%左右。淀粉葛根素同步提取最佳工艺为:鲜葛第一次提取淀粉时同步添加2%的D101大孔树脂,吸附时间90 min,吸附搅拌速度150 r/min,解吸附乙醇浓度60%,解吸附剂体积为废水的30%,解吸2次,每次解吸附时间30 min。通过同步提取工艺,实现淀粉提取时第一次洗涤废水中葛根素回收率达85%以上,并可有效缩短提取时间,避免在葛粉沉降过程中废水经过微生物发酵、物理化学反应等影响葛根素回收效果,为葛根资源的高值化利用提供科学依据。 Puerarin is the main active ingredient of Pueraria thomsonii Benth,but in the process of producing pueraria starch,a large amount of puerarin is lost with the processing wastewater,resulting in resource waste.Simultaneous recovery of puerarin from wastewater during the extraction of pueraria starch is an important way to achieve high-value utilization of pueraria resources.The article used high-performance liquid chromatography to analyze the distribution pattern of puerarin content in different parts of pueraria and in the wastewater of pueraria starch extraction process.Single factor experiments and orthogonal experimental designs were used to optimize the simultaneous extraction process of pueraria starch and puerarin.The results showed that over 80%of puerarin was concentrated in pueraria bark,and the content of puerarin in the first starch extraction wastewater accounted for about 70%of the total wastewater.The optimal process for simultaneous extraction of starch and puerarin from fresh pueraria is to simultaneously add 2%D101 macroporous resin during the first extraction of starch,with an adsorption time of 90 minutes,an adsorption stirring speed of 150 r/min,a desorption ethanol concentration of 60%,a desorption agent volume of 30%wastewater,desorption twice,and a desorption time of 30 minutes each time.By using simultaneous extraction technology,the recovery rate of puerarin in the first washing wastewater during starch extraction can reach over 85%,and the extraction time can be effectively shortened to avoid the impact of microbial fermentation,physical and chemical reactions,etc.on the recovery efficiency of puerarin in the wastewater during the settling process of pueraria starch.This provides a scientific basis for the high-value utilization of pueraria resources.
