摘要
目的基于网络药理学探讨补阳还五汤治疗新型冠状病毒肺炎继发肺纤维化的作用机制。方法利用中药系统药理学分析平台(TCMSP)数据库、相关文献筛选出补阳还五汤的活性成分及相关靶点,在GeneCards和在线人类孟德尔遗传(OMIM)数据库中检索新型冠状病毒肺炎与肺纤维化的疾病靶点,取三者的交集靶点。利用Cytoscape 3.7.2软件、String在线网站分别构建“药物-活性成分-疾病-靶点”网络和蛋白互作网络(PPI)。最后,利用R x644.1.2软件进行富集分析。结果筛选得到106个补阳还五汤的活性成分和70个药物-疾病共同靶点。富集分析结果涉及细胞因子及其受体配体活性等方面,其作用机制可能与白细胞介素-17(IL-17)、Toll样受体、肿瘤坏死因子(TNF)等信号通路有密切关系。结论补阳还五汤可通过多成分、多靶点、多信号通路发挥治疗新型冠状病毒肺炎继发肺纤维化的作用。
Objective To explore the mechanism of Buyang Huanwu decoction in the treatment of pulmonary fibrosis secondary to COVID-19 based on network pharmacology.Methods TCMSP database and relevant literature were used to screen out the active ingredients and related targets of Buyang Huanwu decoction.The disease targets of COVID-19 and pulmonary fibrosis were searched in GeneCards and OMIM databases,and the intersection targets of the three were taken.Cytoscape 3.7.2 software and String online website were used to construct the drug-active ingredient-disease-target network and protein interaction network(PPI)respectively.Finally,enrichment analysis was performed using R x644.1.2 software.Results A total of 106 active ingredients and 70 drug-disease common targets of Buyang Huanwu decoction were screened.The results of enrichment analysis involved the activities of cytokines and their receptor ligands,and its mechanism may be closely related to IL-17,Toll-like receptors,TNF and other signaling pathways.Conclusion Buyang Huanwu decoction can play a role in the treatment of pulmonary fibrosis secondary to COVID-19 through multi-components,multi-targets and multi-signaling pathways.
